Adaption to chronic hypercapnia drives time- and site-dependent changes in brainstem cardiorespiratory nuclei.


ABSTRACT: Traditional mechanisms of respiratory neuroplasticity to not fully describe the temporal pattern of ventilatory adaptations to chronic hypercapnia. To gain insights into novel mechanisms that may underly such adaptations, we performed bulk-tissue RNA sequencing from three key brainstem cardiorespiratory site involved in acute chemosensitivity (the retrotrapezoid nucleus; RTN), respiratory rhythm generation (the ventral respiratory column, VRC), and peripheral information integration (the nucleus tractus solitarius, NTS) during three different time-points of acclimatization to chronic hypercapnia (3 hours (h), 24h, and 7 days (d)). We hypothesized that chronic hypercapnia induces both time- and region-dependent transcriptional changes that may partially underly ventilatory and chemoreceptive adaptations.
