Human airway eptithelial cell subsets exhibit distinct transcriptional profiles in response to IFN-gamma derived from influenza specific CD8 T cells.

ABSTRACT: To evaluate the impact of IFNγ secretion by influenza-specific CD8⁺ T cells on human lung epithelial cells, we stimulated total primary human lung cells with influenza peptides in the presence or absence of an IFNγ-blocking antibody. Following stimulation, epithelial cells (CD45⁻, CD31⁻, CD326⁺) were isolated by FACS and analyzed by single-cell RNA sequencing. Three stimulation conditions were compared including unstimulated, stimulated with influenza peptides plus isotype control antibody, and stimulated with influenza peptides plus anti-IFNg antibody.
