Stage-specific microRNA profiling of FACS-sorted Math1-positive commissural neurons in developing chick embryos

ABSTRACT: This study aims to systematically identify miRNAs and characterize microRNA (miRNA) expression in a specific population of Math1-positive commissural neurons in the developing chick spinal cords. To achieve this, chick embryos were electroporated with constructs encoding a Venus-tagged, enhancer-driven Math1, followed by FACS isolation of Venus-Math1-positive neurons from developing spinal cords at pre-crossing (HH18–20) stages. By performing miRNA sequencing on these sorted populations, the study seeks to uncover stage-specific miRNA expression signatures that may play critical roles in the differentiation, identity, and maturation of Math1-positive commissural neurons during spinal cord development.
