Comparison of gene expression between mouse lung resident MSC subpopulations.

ABSTRACT: Lung resident MSCs were isolated from the Tbx4-rtTA/TetO-Cre/mT-mG reporter mice at 3 months of age, in which embryonic lung mesenchymal cells are genetically marked by doxycycline induction from embryonic day 6.5 to postnatal day 1. The cultured MSCs at passage 2 were sorted by FACS and total RNA was isolated. Samples were from different isolations.

Project description:Transgenic mice (Scgb1a1-rtTA/[tetO]-KRAS.G12D/Nkx2-1+/-) develop mucinous lung tumors. Gene expression analysis was performed using mRNAs from the whole lungs of the mice compared to that of the control mice. We used microarrays to detail the global program of gene expression underlying development of mucinous lung tumors and identified distinct classes of up-regulated genes during this process. mRNAs were isolated from the whole lungs of the transgenic mice (Scgb1a1-rtTA/[tetO]-KRAS.G12D/Nkx2-1+/-) after 2 months doxycycline administration and hybridized on Affymetrix microarrays. We sought to identify genes induced in mucinous lung tumors.

Project description:Lung resident MSCs were isolated from a deidentified donor. After ~14-day culture, individual colonies were isolated using a cloning ring approach. Individual colonies were separately expanded and the cells at passage 4 were collected for total RNA isolation.

Effect of Misexpression of Sox9 on Gastric Differentiation

Project description:To determine if Sox9 was sufficient to drive mucous differentiation in the gastric epithelium, we bred mice carrying alleles for ROSA26rtTA.IRES.EGFP, TetO-Sox9, and Fgf20Cre.GFP. In these mice, cells expressing Cre will induce rtTA from the ROSA26rtTA.IRES.EGFP allele. To explore transcriptional changes following adult misexpression of Sox9, we isolated RNA from the corpus of Fgf20Cre.GFP/Cre.GFP; ROSA26rtTA.IRES.EGFP/rtTA.IRES.EGFP; TetO-Sox9 pup (misexpression animals) and ROSA26rtTA.IRES.EGFP/rtTA.IRES.EGFP; TetO-Sox9 pup (control) animals and performed RNA-sequencing.

Single cell RNA-seq analysis of Tbx4+ and Tbx4- mouse lung mesenchymal cells

Project description:Purpose: The goals of this study are to analyze Tbx4 lineage+ and Tbx4 lineage- cells sorted from Tbx4cre;RosaTm mouse lung. Methods: Tbx4 lineage+ and Tbx4 lineage- cells within epcam+ CD31- CD45- 7AAD- cells were sorted from Tbx4cre;RosaTm mouse lung, and run on the C1 chip (Fluidigm). Cell capture, lysis, reverse transcription and cDNA amplification was performed on the C1 IFC for mRNA-seq on Fluidigm C1 Single-Cell Auto Prep System following the manufacturer’s protocol. Results: C1 single cell RNA-seq platform captured and sequenced 54 Tbx4 lineage+ cells and 39 Tbx4 lineage - cells. Conclusions: Our study analyzed transcriptomes of Tbx4 lineage+ and Tbx4 lineage- lung mesenchymal cells. This study provides a framework for the application of transcriptome profiling towards characterization of differentiated genes Tbx4 lineage+ and Tbx4 lineage - cells.

RNA-seq analysis of Mst1/2 deleted bronchiolar epithelial cells from adult mouse lungs

Project description:Mst1 and Mst2 were conditionally deleted from non-ciliated bronchiolar epithelial cells in the mature lung. Bronchiolar epithelial cells from control and Mst1/2 deleted mice were isolated by cell sorting and used for RNA-seq analysis. Scgb1a1-rtTA/tetO-Cre/Mst1;2-flx/flx (Mst1/2 D/D) mice were generated to conditionally delete Mst1 and Mst2 from non-ciliated, secretory bronchiolar epithelial cells. Adult mice were maintained on doxycycline food for 16 days to induce deletion of Mst1/2. Lin-/CD326+/CD24-intermediate cells were isolated by fluorescence cell sorting to enrich for the targeted airway epithelial cells. mRNA isolated from Lin-/CD326+/CD24-intermediate cells from control and Mst1/2 D/D mice was pooled and analyzed by RNA-seq to identify transcriptional changes following deletion of Mst1 and Mst2 from mature lung bronchiolar epithelial cells.
