Effect of PRLX-93936 treatment on gene expression of PANC-1 cells

ABSTRACT: PRLX-93936 is an optimized derivative of erastin. To determine the downstream effects of PRLX-93936 on gene expression, we performed mRNAseq of PANC-1 cells treated with the drug or vehicle control.

Project description:cGMP is well-known secound messanger involved in vascular homeostasis. However little is known the effect of cGMP inducer on mRNA expression in cancer cells. We performed microarrays to revealed the transcriptional change of a human pancreatic ductal adenocarcinoma (PDCA) cell line (Panc -1) to cGMP inducer Bay41-2272 in order to provide insight into impact of cGMP induction on Panc-1 cells. Panc-1 Human PDCA cells were treated with vehicle (DMSO) or 5 Î¼M Bay41-2272 for 48 h and total RNA was extracted by using TRIzol Reagent. The microarray analysis was conducted on the Panc-1 cells expressing by using Agilent Microarray.

Project description:Analysis of differentially expressing genes in whole genome wide analysis of aptamer SQ2 positive cells (Capan-1, Panc-1, Panc-1+ve) and SQ2 negative cells (Panc-1-ve and HPDE) Panc-1 +ve and Panc-1-ve cell lines were generated from Panc-1 cell line based upon its hetergenous binding to aptamer SQ2. Detailed procedure of generation of these cell lines are described in Pooja Dua, Hye Suk Kang, Seung-Mo Hong, Ming-Sound Tsao, Soyoun Kim, and Dong-ki Lee. 2012 Alkaline Phosphatase ALPPL2 is a novel pancreatic carcinoma-associated protein. Cancer Research A five chip study using total RNA recovered from Capan-1, Panc-1, Panc-1+ve, Panc-1-ve and HPDE cells. Each chip measures 45,033 genes with three 60 mer probe pairs per target.

Expression data by BRD7552 treatment in PANC-1 cells

Project description:Three master regulatory transcription factors Pdx1, MafA and Ngn3 have the ability to transdifferentiate pancreatic acinar cells to insulin-producing beta cells in mice. BRD7552 was identified as a small-molecule inducer that can upregulate the expression of Pdx1 in PANC-1 cells by high-throughput qPCR screening. We used microarrays to illustrate the mechnism of BRD7552 for PDX1 and insulin induction. PANC-1 cells were treated with BRD7552 at 5uM, the mininal concentration for the maximal induction of Pdx1. The RNA was isloated at two time points 6-hour and 72-hour.

The effect of FOXO3 knockdown and PDHA1 knockdown on PDAC cell line Panc-1

Project description:Little is known about the role of FOXO3 and PDHA1 in PDAC. We performed microarrays to revealed the transcriptional change of a human pancreatic ductal adenocarcinoma (PDCA) cell line (Panc -1) to scr-siRNA, FOXO3-siRNA and PDHA1-siRNA in order to provide insight into the role of PDHA1 and FOXO3 in Panc-1 cells. Panc-1 Human PDCA cells were treated with scr-siRNA, FOXO3-siRNA and PDHA1-siRNA for 48 h and total RNA was extracted by using TRIzol Reagent. The microarray analysis was conducted on the Panc-1 cells expressing by using Agilent Microarray.

Gene expression analysis in Panc-1 cells in response to treatment with Gli3T, a dominant-negative repressor of Gli

Project description:Pancreatic Ductal Adenocarcinoma (PDAC) is one of the most aggressive human malignancies. In our studies, we find that the Gli transcription factors are required for Kras initiation of pancreatic tumorigenesis. In order to identify the downstream transcriptional targets of Gli in PDAC, we conducted gene expression analysis using Gli3T, a transcriptional repressor of Gli. In this data set we include the expression data from 6 samples with three of them expressing a control vector and another three expressing Gli3T Gli3T with IRES-GFP, or an empty vector was ectopically expressed in Panc-1 cells using lipofectamine transfection. GFP positive cells were sorted 24 hours post transfection and RNA was isolated from the cells using Qiagen RNAeasy kit. Affymetrix gene expression array was conducted on the Panc-1 cells expressing the control vector and cells expressing Gli3T. Relative gene expression between the two sample sets was analyzed. Three samples were analyzed for each set.
