Project description:Enterobacter sp. SA187 is a plant growth-promoting bacterium (PGPB) that promotes growth of the crop plant alfalfa under saline irrigation and desert farming conditions. SA187 also enhances salt tolerance of the model plant Arabidopsis thaliana under in vitro conditions. In the present study, we used a transcriptomic approach to elucidate the mechanisms underlying plant growth promotion by SA187 under salt stress. Compared to free-living SA187, a massive metabolic reprogramming of SA187 occurs upon association with Arabidopsis. This effect was largely independent of the plant growth condition (non-salt or salt stress). Our data revealed pronounced changes in gene expression of proteins involved in cell signaling, chemotaxis, flagella biosynthesis, quorum sensing and biofilm formation. Also, upon plant interaction, a complete reprograming of nutrients acquisition and the central carbon metabolism of SA187 was observed. Moreover, in accordance with the previously identified role of bacterially produced 2-keto-4-methylthiobutyric acid (KMBA) in mediating salt stress tolerance, the sulfur metabolism of SA187 was strongly induced. Overall, our results give a deep insight into the metabolic and signaling pathways involved in the transition from free-living to a plant-associated PGPB life style of SA187.

Project description:Recent studies have shown that several plant species require microbial associations for stress tolerance and survival. In this work, we show that the desert endophytic bacterium Enterobacter sp. SA187 enhances yield and biomass of alfalfa in field trials, revealing a high potential for improving desert agriculture. To understand the underlying molecular mechanisms, we studied SA187 interaction with Arabidopsis thaliana. SA187 colonized surface and inner tissues of Arabidopsis roots and shoots and conferred tolerance to salt and osmotic stresses. Transcriptome, genetic and pharmacological studies revealed that the ethylene signaling pathway plays a key role in mediating SA187-triggered abiotic stress tolerance to plants. While plant ethylene production is not required, our data suggest that SA187 induces abiotic stress tolerance by bacterial production of 2-keto-4-methylthiobutyric acid (KMBA), known be converted into ethylene in planta. These results reveal a part of the complex molecular communication process during beneficial plant-microbe interactions and unravel an important role of ethylene in protecting plants under abiotic stress conditions.

SA187 induces thermotolerance of crops in a sustainable manner. Microbiome diversity of wheat plants is positively influenced by SA187 in open field agriculture, indicating that beneficial microbes can be a powerful tool to enhance agriculture in open field agriculture.

Project description:High light stress in subtropical and tropical regions strongly limits agricultural production due to photo-oxidative damage, decreased growth and yield. Here, we investigated whether beneficial microbes can protect plants under high light stress. We show that Enterobacter sp. SA187 (SA187) assists Arabidopsis in maintaining growth under high light stress, reducing the accumulation of reactive oxygen species (ROS) and maintaining photosynthesis. Under high light stress, SA187 induces dynamic transcriptional changes related to a fortified iron metabolism and redox system in Arabidopsis. A genetic analysis shows that SA187-induced plant high light stress tolerance is mediated by ethylene signaling via the transcription factor EIN3 to enhance iron metabolism. In summary, we show that Arabidopsis interaction with SA187 results in sustained photosynthesis under high light stress suggesting that beneficial microbes could be an effective and inexpensive means for enhancing high light stress tolerance in crops.

Project description:Enterobacter sp. SA187 is a facultative endophytic bacterium conferring multi-abiotic stress tolerance to various plant hosts. Upon interaction with plant tissues, a significant proportion of the typically yellow SA187 lose pigmentation. This phenotypic shift becomes more prominent with extended host plant colonization and under stress conditions, such as salinity. To explore the underlying mechanisms and ecological significance of this variation, we employed genome sequencing, comparative genomics, transcriptomics, and metabolic characterization. In all white SA187 variants, weidentified consistent point mutations in the rpoS gene, which encodes a global regulatory sigma factor. These rpoS loss-of-function mutations lead to alterations in gene regulation, affecting growth, morphology, biofilm formation, motility, oxidative stress responses and carotenoid production. Notably, the rpoS mutants demonstrated enhanced adaptability from a free living to an endophytic life style. Whereas the desert soil is characterized by highly alkaline conditions, the apoplast of the host plant is an acidic environment accompanied with the availability of distinct carbon sources. RpoS mutants allow life in the acidic and sucrose-rich apoplastic compartment, underscoring the role of genetic variation in bacterial adaptation to colonize plants.

Project description:Background and aimsMineral elements have many essential and beneficial functions in plants. Phosphorus (P) deficiency can result in changes in the ionomes of plant organs. The aims of this study were to characterize the effects of P supply on the ionomes of shoots and roots, and to identify chromosomal quantitative trait loci (QTLs) for shoot and root ionomic traits, as well as those affecting the partitioning of mineral elements between shoot and root in Brassica napus grown with contrasting P supplies.MethodsShoot and root concentrations of 11 mineral elements (B, Ca, Cu, Fe, K, Mg, Mn, Na, P, S and Zn) were investigated by inductively coupled plasma optical emission spectrometry (ICP-OES) in a Brassica napus double haploid population grown at an optimal (OP) and a low phosphorus supply (LP) in an agar system. Shoot, root and plant contents, and the partitioning of mineral elements between shoot and root were calculated.Key resultsThe tissue concentrations of B, Ca, Cu, K, Mg, Mn, Na, P and Zn were reduced by P starvation, while the concentration of Fe was increased by P starvation in the BnaTNDH population. A total of 133 and 123 QTLs for shoot and root ionomic traits were identified at OP and LP, respectively. A major QTL cluster on chromosome C07 had a significant effect on shoot Mg and S concentrations at LP and was narrowed down to a 2.1 Mb region using an advanced backcross population.ConclusionsThe tissue concentration and partitioning of each mineral element was affected differently by P starvation. There was a significant difference in mineral element composition between shoots and roots. Identification of the genes underlying these QTLs will enhance our understanding of processes affecting the uptake and partitioning of mineral elements in Brassica napus.

Project description:Plant-soil feedback studies attempt to understand the interplay between composition of plant and soil microbial communities. A growing body of literature suggests that plant species can coexist when they interact with a subset of the soil microbial community that impacts plant performance. Most studies focus on the microbial community in the soil rhizosphere; therefore, the degree to which the bacterial community within plant roots (root-endophytic compartment) influences plant-microbe interactions remains relatively unknown. To determine if there is an interaction between conspecific vs heterospecific soil microbes and plant performance, we sequenced root-endophytic bacterial communities of five tallgrass-prairie plant species, each reciprocally grown with soil microbes from each hosts' soil rhizosphere. We found evidence of plant-soil feedbacks for some pairs of plant hosts; however, the strength and direction of feedbacks varied substantially across plant species pairs-from positive to negative feedbacks. Additionally, each plant species harbored a unique subset of root-endophytic bacteria. Conspecifics that hosted similar bacterial communities were more similar in biomass than individuals that hosted different bacterial communities, suggesting an important functional link between root-endophytic bacterial community composition and plant fitness. Our findings suggest a connection between an understudied component of the root-endophytic microbiome and plant performance, which may have important implications in understanding plant community composition and coexistence.
