Single-cell transcriptome profiling of tracheal epithelium in basal cell-specific Mettl3 conditional knockout mice

ABSTRACT: The airway epithelium plays critical roles in respiratory function and is implicated in various pulmonary diseases. To investigate the role of the RNA methyltransferase Mettl3 in airway basal cell homeostasis, we generated a comprehensive single-cell RNA sequencing atlas of tracheal epithelial cells from Mettl3 conditional knockout (M3-CKO) mice and littermate controls. Our dataset specifically focuses on Krt5-lineage traced basal cells, comparing tdTomato+ (traced) and tdTomato- (non-traced) populations from both genotypes.We present a comprehensive single-cell RNA-sequencing atlas of tracheas, comprising diverse populations of cells, epithelial cell, myeloid cells, B lymphocytes, T lymphocytes, endothelial cells, fibroblasts and glandular mucous cells.
