Epithelial Edar is Responsible for Dental Anomalies in HED


ABSTRACT: In this study, we generated epithelial-specific Edar knockout mouse models and reported that the epithelial Edar is responsible for ectodermal dysplasia, determines tooth size and influences tooth morphology and number. Ko is from K14cre;Edarfl/fl mice and ctrl is from Edarfl/fl mice. Single-cell RNA sequencing revealed that impaired differentiation from preameloblasts to early ameloblasts may underlie the observed enamel defect. Taken together, these findings reveal a critical role of epithelial EDAR in tooth development and, for the first time, provide evidence that its downregulated expression can lead to enamel formation defects, offering new insights into the pathogenic mechanisms of EDAR-related HED.
