MRNA sequencing data of hepatic macrophages (Kupffer cells) under endotoxin tolerance, LPS-induced activation, and control conditions

ABSTRACT: Endotoxin tolerance is an important adaptive response of macrophages following repeated exposure to lipopolysaccharide (LPS), leading to a reduced pro-inflammatory response. To investigate the transcriptional changes associated with in vitro LPS-induced tolerance in liver-resident macrophages, we isolated primary Kupffer cells from wild-type C57BL/6 mice and subjected them to distinct LPS stimulation protocols in vitro. Three groups were analyzed: untreated control cells (Ctrl), cells stimulated once with LPS at the second time point (2nd), and cells stimulated with LPS at both the first and second time points (Tol). Total RNA was extracted from each group, and 3'-end mRNA sequencing (QuantSeq 3’ mRNA-Seq) was performed. This dataset provides insight into the transcriptional programs underlying LPS-induced tolerance in Kupffer cells.
