Notch signaling is required for induction of an innate T cell development program in human thymus

Project description:CD4 CD8 double positive (DP) thymocytes progressively shut down Notch signaling after β-selection allowing progression of the DP program and positive selection of conventional CD4 and CD8 single positive T cells. In contrast, innate CD8+ T cells are selected from DP precursors shortly after β-selection. Here, we investigated whether notch signaling is required for the initiation of the innate T cell developmental program upon TCR engagement. We show that, after β-selection, Notch activation limits progression to late DP cells and favors the development of TCRαβ/CD3 expressing CD8dim early DP progenitors. TCR engagement in these Notch dependent TCRαβ+ early DP precursors does not induce prominent apoptosis, but rather induces T-bet expression by an mTOR-dependent mechanism. Our findings indicate that Notch signaling drives the launch of an innate effector program in response to TCR agonist selection in the thymus.

Project description:The murine thymus produces discrete Î³Î´ T cell subsets making either IFN-Î³ or IL-17, but the role of the TCR in this developmental process remains controversial. Here we generated a non-transgenic and polyclonal model of reduced TCR expression and signal strength selectively on Î³Î´ T cells. Mice haploinsufficient for both CD3Î³ and CD3Î´ (CD3DH) showed normal Î±Î² thymocyte subsets but specific defects in Î³Î´ T cell development, namely impaired differentiation of IL-17-producing embryonic VÎ³6+ (but not adult VÎ³4+) Î³Î´ T cells and a marked depletion of IFN-Î³-producing CD122+ NK1.1+ (VÎ³1-biased) Î³Î´ T cells throughout life. As result, adult CD3DH mice showed defective peripheral IFN-Î³ responses and were resistant to experimental cerebral malaria. Thus, strong TCR signaling is required within specific developmental windows with distinct VÎ³ usage and differential cytokine production by effector Î³Î´ T cell subsets. Liver samples from 14 fetuses were obtained at gestational week 8-12. Adult liver samples were collected from 50 organ donors who had met accidental death and 42 liver samples from patients undergoing liver resection due to malignant tumors, most commonly from patients with metastatic colon cancers. Liver biopsies from these patients were collected from 'healthy' tissue that showed no visible pathological changes compared to the adjacent tumor.
