B cell gene expression from allergen-encapsulating nanoparticle-treated allergic mice

ABSTRACT: Polymeric allergen-loaded nanoparticles were used as a treatment for a murine model of food allergies. To study changes in B cell gene expression due nanoparticle treatment, bulk RNA sequencing of B cells from the peyer’s patches (PP) and spleen was done after treatment.

Mucosal immune response to persistent Mycobacterium avium ssp. Paratuberculosis (MAP) infection in bovine gut-associated lymphoid tissues

Project description:Chronic enteric Mycobacterium avium subsp. paratuberculosis (MAP) infections are endemic in cattle worldwide but disease management is thwarted by a lack of therapeutics to prevent or control infection. A better understanding of the host-pathogen interactions occurring in Peyer’s patches (PP), the portal of MAP entry and site of infection, is required to develop effective vaccines. The ruminant small intestine possesses two structurally and functionally distinct PP (discrete [dPP] found in jejunum and continuous [cPP] found in ileum). Using RNA-Seq, we investigated the long-term outcome of persistent MAP infection in dPP compared to cPP in neonatal calves using surgically isolated intestinal segments containing draining mesenteric lymph nodes (MLN) and Peyer’s Patches (PP). At 12 months post-infection, MAP persisted at a detectable level in cPP (n=4), but was controlled in dPP (n=5). This study demonstrates a marked regional difference in both persistence of MAP infection and the mucosal immune responses that occur in dPP and cPP following infection.

Neutrophil extracellular traps melt Peyer’s patches following stroke and myocardial infarction

Project description:Sterile tissue injury after stroke causes lymphocyte contraction in lymphoid tissues and may decrease circulating IgA-levels. Intestinal Peyer’s patches (PP) harbor large numbers of IgA+ B cell precursors and plasma cells. Whether and how tissue injury triggers PP-B cell death, thereby mediating IgA-loss, is unknown. We found decreased circulating IgA levels in stroke and myocardial infarction patients. Experimental stroke and myocardial infarction in mice phenocopied the human situation. Decreased plasma and fecal IgA were accompanied by rapid and macroscopic shrinkage of PP caused by substantial losses of PP-resident IgA+ precursors and plasma cells in mice. Tissue injury induced neutrophil activation endowed with the release of toxic neutrophil extracellular traps (NETs). Antibody-mediated or genetically- induced neutrophil loss, digestion of NETs, or inhibition of their release by the Gasdermin D blockade completely prevented lymphocyte loss and PP shrinkage. We also identified NETs in the plasma of stroke and myocardial infarction patients. Hence, tissue injury induces systemic NET-release, which might be targeted to maintain immune homeostasis at mucosal barriers.

Project description:Transcriptome analyses were done comparing gene regulation of TFH and TFR cells isolated from Peyer’s Patches (PP) and immunized peripheral lymph nodes (pLN) revealing many regulatory patterns common to all follicular cells. However, in contrast to TFH cells, the kinetic but also the extent of regulation regarding many genes was attenuated in pLN TFR cells when compared to those of PP. Additionally, PP but not pLN TFR cells were largely unresponsive to IL2 and expressed Il4 as well as Il21. Together with fundamental differences in gene expression that were found between cells of both compartments this emphasizes specific adaptations of follicular T cell functions to their micro-milieu.

Project description:We combine single-cell RNA sequencing (scRNA-seq) with physiological and genetic perturbations to dissect the cellular circuit of type 2 intestinal inflammation. To uncover key responding cellular components, we profiled individual immune cells in the lamina propria (LP) and Peyer’s patches (PPs), regions of the small intestine enriched for immune cells, in homeostasis and in an intestinal type 2 inflammatory model. We obtained 36,797 and 21,270 high-quality cells passing initial filtering from PP and LP regions, respectively.

Expression data from murine follicular dendritic cells

Project description:Germinal centers (GCs) are clusters of activated B cells built on stromal cells known as follicular dendritic cells (FDCs). In the Peyer’s patches (PPs), GCs are chronically induced by bacteria and are the major sites for generation of gut IgA immune responses. Whether FDCs directly contribute to the IgA production in PP GCs is unknown. To investigate the role FDCs in gut immune system, we examined comprehensive gene profiles of FDCs purified from PPs or perypheral lymph nodes (pLNs) with or without immunization. We also tried to reconstitute the PP FDC signature in vitro by pulsed or continuous stimulation of pLN FDCs through TLRs, RARs or simultaneously through TLRs and RARs.
