Project description:The integration of extrinsic signaling with cell-intrinsic transcription factors can direct progenitor cells to differentiate into distinct cell fates.In the developingDrosophilaeye, differentiation of photoreceptors R1-R7 requires EGFR signaling mediated by the transcription factor Pointed, and our single-cell RNA-Seq analysis shows that the same photoreceptors require the eye-specific transcription factor Glass. We discovered that ectopic expression of Glass and activation of EGFR signaling synergistically induce neuronal gene expression in the wing disc in a Pointed-dependent manner. Targeted DamID reveals that Glass and Pointed share many binding sites in the genome of developing photoreceptors. Comparison with transcriptomic data shows that Pointed and Glass induce photoreceptor differentiation through intermediate transcription factors, including the redundant homologues Scratch and Scrape, as well as directly activating neuronal genes. Our data reveal synergistic activation of a multi-layered transcriptional network as the mechanism by which EGFR signaling induces neuronal identity in Glass-expressing cells.

Project description:Following sex determination, XY and XX gonads develop into a testis and an ovary, respectively. Depending on the sex of the gonad, resident germ cells will subsequently be committed to either spermatogenesis or oogenesis. In this study we took advantage of the Wv/Wv mouse genetic model, in which gonads are almost devoided of germ cells, to uncover gene expression underlying fetal germ cell development. Male and Female gonads were collected at 12, 14 and 16 days post coitum (dpc) from wild-type and Wv/Wv mouse embryos. Total RNAs were extracted and hybridized on Affymetrix microarrays. Expression signals from male and female gonads or from wild-type and mutant gonads were compared to identify sexually dimorphic genes as well as genes expressed in germ cell during fetal gonad development.

Project description:We performed CAGE-Seq on dissected ovaries and testes as well as female and male carcasses of two species of Drosophila (D. melanogaster and D. pseudoobscura). These data are used to map transcription start sites. Project description:In this study, we attempt to characterize the transcriptomic profile of the Asian seabass gonads at various developmental stages. The protandric Asian seabass or barramundi (Lates calcarifer) typically matures as a male at approximately 2M-bM-^@M-^S4 years of age and then changes sex to a female in later years. For this experiment, Asian seabass of several ages were collected from the Marine Aquaculture Center of the Agri-Food & Veterinary Authority of Singapore and from farms around Singapore. There were no treatments carried out in this experiment. The gonads were examined by histology and classified according to sexual maturation status as described by Guiguen and colleagues (Guiguen et al. Environmental Biology of Fishes, 1994). Altogether, we analyzed 22 gonadal samples that could be classified into six different types of gonads. Total 22 samples: Adult Ovaries (F3-stage; 5 years old fish) : 4 Adult Testes (M3-stage; 5 years old fish) : 4 Early Testes (M3-stage; 8-9 months old fish) : 3 Early Transforming Gonads (>2 years old fish) : 3 Late Transforming Gonads (>2 years old fish) : 4 Undifferentiated Gonads (4.5 months old fish) : 4

Project description:Adult ovarian tissues or biopsies isolated from patients prior to chemotherapy or irradiation can reconstitute ovarian functions when transplanted either in the abdomen or subcutaneously. This approach avoids invasive surgery and potential risks associated with internal procedures. We investigated whether functional ovaries could develop subcutaneously from early E12.5 fetal gonads without entering meiosis in mouse model. Unexpectedly, the subcutaneously transplanted fetal gonads failed to undergo folliculogenesis, showing meiotic deficiencies, DNA repairdefects, and elevated apoptosis. Thes meiotic defects were partly due to variations in temperature and oxygen concentration. However, in vitro culture of the gonads at 37C allowed completion of meiotic prophase I. After subcutaneous transplantation, the in vitro cultured E12.5 gonads successfully underwent folliculogenesis, restoring endocrine functions. This suggests potential for using pluripotent stem cell technologies to rejuvenate ovarioids from fetal gonad-like cells, enhancing endocrine recovery and health span.
