Comparative Analysis of microRNA Expression in Serum and Plasma in Patients Screened for BRCA1/2 Mutations, dataset 2

ABSTRACT: Serum samples from 94 healthy women were analyzed using a custom qPCR panel comprising 182 miRNAs. Since this experiment is part of a broader study on miRnome alterations associated with BRCA mutations or BRCA-related cancers (particularly ovarian and breast cancer), 37 of the analyzed samples came from patients with BRCA1 mutations and 19 from BRCA2 mutation carriers. This analysis was preceded by sequencing-based quantification on the same set of the samples, the results of which are submitted separately to GEO under accession number GSE299787. The generation of this dataset was supported by The Gray Foundation grant “Circulating microRNAs for assessment of risk beyond the BRCA genes and early detection of breast cancer in high-risk families” awarded to Dipanjan Chowdhury, and by the National Science Centre (Poland) OPUS grant “Predictive Potential of Circulating MicroRNA Biomarkers in Patients with High Familial or Genetic Risk of Cancer” (2023/49/B/NZ5/03835) awarded to Wojciech Fendler.

Project description:Paired serum and plasma samples from 23 patients were analyzed using a custom qPCR panel comprising 182 miRNAs. Project description:This dataset was created in order to evaluate the concordance of miRNA expression between serum and plasma in humans. miRNA expression was quantified in both biological materials using sequencing. Differential expression and correlations analysis were used to evaluate similarities and differences between miRNAs abundance in plasma and serum. Concurrently, miRNA quantification in the subset of these samples was performed by qPCR, the results of which belong to the separate submission to GEO. Project description:This dataset consists of miRNA expression in serum of 69 women. This data is the result of remapping raw sequencing FASTQ files being a part of the GSE226445 dataset and the SRA Bioproject PRJNA898621. MicroRNA abundance was quantified by sequencing. Data was analyzed together with plasma samples of those same patients with the aim of assessing the impact of blood processing steps on quantified miRNA abundance. Plasma data belongs to a separate submission (accession will be added when issued). Serum samples of remaining 67 women have been already submitted to GEO in GSE226445 dataset and to SRA as Bioproject PRJNA898621, while counts generated in re-mapping for this project are deposited as GSE299846 data set. Cholesin Receptor Signalling is Active in Cardiovascular System-Associated Adipose Tissue and Correlates with SGLT2i Treatment in Patients with Diabetes

Project description:Objective: To identify the cholesin receptor-associated genes and clinical factors linked with their expression in cardiovascular system and associated adipose tissues. Research Design and Methods: Right cardiac auricle, aortic wall, saphenous vein and adipose tissue (periaortic-PAT, epicardial-EAT, thymic-TAT) samples were collected during coronary-artery bypass grafting. Results: We included 46 participants [37 male, 23 with type 2 diabetes, median age 68.50 (Q1-Q3: 63.00-72.00) years, BMI 28.39 (26.06-31.49) kg/m2]. GPR146 expression in adipose tissues significantly correlated with BMI, c-peptide, total cholesterol and LDL concentrations. Selected metabolic pathways were significantly and positively enriched in GPR146-dependent manner. GPR146-coexpressed genes contained key regulators of lipid metabolism, and correlated positively with LDL, HDL and total cholesterol concentrations. SGLT2i treatment mirrored GPR146-associated signature in EAT, suggesting potential impact of SGLTi on cholesin-GPR146 network. Conclusions: GPR146 expression is associated with serum lipids and metabolically-relevant transcriptomic changes in adipose tissue, in EAT similar to iSGLT2-associated ones. Comparative multi-omic analysis of fallopian tube fimbria from BRCA mutation carriers and control women

Project description:Integrated transcriptomic, epigenomic, and proteomic analysis of histologically normal fallopian tubes (FTs) fimbria from BRCA mutant carriers and controls. This study was undertaken to test the hypothesis that there are molecular alterations in FTs from BRCA mutant carriers that contribute to field cancerization and high-grade serous risk. We address the limitations of prior molecular profiling studies of BRCA mutant FTs by controlling for covariates known to influence the molecular and cellular landscape of the FT.
