Exosomal miRNA Profiling in Plasma and Cerebrospinal Fluid of DOCA-Salt-Induced Hypertensive and Control Mice


ABSTRACT: This study aimed to profile exosomal miRNAs in both plasma and cerebrospinal fluid (CSF) from male and female mice subjected to DOCA-salt treatment—a widely used model for inducing hypertension—and from their respective baseline controls. Exosomes were isolated using size exclusion chromatography (SEC), followed by small RNA extraction and quality control. Exosomal miRNAs were sequenced using the QIAseq miRNA Library Kit (Qiagen) on the Illumina NovaSeq 6000 platform. Libraries incorporated unique molecular identifiers (UMIs) to mitigate PCR bias. High-throughput sequencing data were analyzed for quality, aligned to multiple databases (miRBase, piRNABank, and Rfam), and normalized using CPM values. Differentially expressed miRNAs were identified and subjected to target prediction and pathway analysis using miRTarBase, GO, and KEGG. These findings may provide insight into the molecular signatures associated with hypertension and sex-specific responses in extracellular vesicle-derived miRNAs.
