Project description:The molecular background of mitochondrial dysfunction in adipose tissue of morbidly obese individuals and bariatric surgery-induced changes in adipose mitochondrial function remain incompletely understood. To evaluate the mechanisms behind the surgery-induced changes and differences between morbidly obese subjects and nonobese controls, we performed a LC-MS/MS proteomics analysis of abdominal subcutaneous (SAT) and visceral adipose tissue samples (VAT) collected from the bariatric surgery, SAT samples collected 6 months after surgery, and control SAT and VAT samples collected from baseline.

Project description:Using RNA isolated from subcutaneous adipose tissue (SAT) and visceral adipose tissue (VAT) samples obtained from control and class I, II and III obese patients undergoing inguinal hernia repair and laparoscopic cholecystectomy, we compared the gene expression profiles between SAT and VAT using microarrays and validated the findings by real-time quantitative PCR. Two-condition experiment, SAT vs. VAT tissue. Biological replicates: 8 SAT replicates, 8 VAT replicates.

Project description:The main objective of this project is to compare the miRNA expression profile of paired visceral adipose tissue and skeletal muscle from obese patients undergoing bariatric surgery. More than 300 miRNAs were identified by Next Generation Sequencing technique in both the visceral adipose tissue and the skeletal muscle of six obese women undergoing bariatric surgery.

Project description:Context: It is not known whether biological differences reported between subcutaneous (SAT) and visceral (VAT) adipose tissue depots underlie the pathogenicity of visceral fat. Objective: We compared SAT and VAT gene expression according to obesity, visceral fat accumulation, insulin resistance and presence of the metabolic syndrome. Design: Subjects were assigned into 4 groups (lean, overweight, obese and obese with metabolic syndrome). Setting: Subjects were recruited at a university hospital. Patients: 32 women were included. Main Outcome Measures: Anthropometric measurements, euglycemic hyperinsulinemic clamps, blood analyses and computed tomography scans were performed and paired samples of SAT and VAT were obtained for DNA microarray-based gene expression profiling.

Epigenome analysis of the human visceral adipose tissue

Project description:Visceral adipose tissue samples were obtained from severely obese individuals that underwent bariatric surgery. The goal of this study was to identify tissue specific methylation QTLs. Whole-transcriptome subcutaneous adipose tissue methylation levels were determined in 71 individuals with a BMI >35 kg/m2. Bisulphite converted DNA from the 71 visceral adipose tissue samples were hybridised to the Illumina Infinium 450k Human Methylation Beadchip.

Selective Reduction of Visceral Adipose Tissue with Injectable Ice Slurry

Project description:Reduction in visceral adipose tissue (VAT) mass reduces body weight and metabolic disease risk in obese patients. However surgical removal of VAT is highly invasive and thus not clinically feasible. We developed an injectable ice slurry for selective reduction of adipose tissue through cryolipolysis. The aim of this study was to investigate safety, feasibility and mechanism of ice slurry-induced cryolipolysis of VAT. Perigonadal VAT in diet-induced obese mice and rats was subjected to slurry or sham treatment. Body weight and blood chemistry were monitored for 56 days post-treatment. Histological analysis and molecular studies were performed to elucidate mechanisms of fat reduction. Treatment of VAT was well tolerated in all animals. Slurry induced adipocyte cell death via selective cryolipolysis; significant weight loss was noted at day 21 post-treatment. RNA sequencing from treated VAT samples showed increased expression of genes involved in inflammation, immune response, collagen biosynthesis and wound healing, and decreased expression of adipokines. This study demonstrates that slurry treatment is safe and effective in inducing cryolipolysis of VAT and subsequent weight loss in rodents. Ice slurry is promising as a minimally-invasive treatment to reduce visceral adipose tissue.
