Oncolytic virus therapy mobilizes tumor-resident CD4+ bystander T cells to restore systemic anti-microbial immunity

ABSTRACT: The library preparation for single-cell RNA sequencing (scRNA-seq) using the BD Rhapsody platform to study SM CD4+ T cells in lung and spleen when undergoing OV-BYTE treatment. Here, we further demonstrate that OV-BYTE delivered dual defense against tumorigenesis and pathogen infections in pre-clinical models. This study gives a transcriptome profiles of SM CD4+ T cells in lung and spleen with administration of OV-BYTE treatment.

Project description:Single-cell RNA sequencing(scRNA-seq) was performed to investigate the differentiation process of steady-state CD4 T cells following treatmernt with IL-25 and IL-33, after initial isolation via FACS. This analysis revealed that steady-state PD-1high CD4 T cell differentiate into patogenic Th2 cells, characterized by the expression of IL-13, IL-5 and amphireguling. These findings suggest that steady-state PD-1high CD4 T cells may contribute to pulmonary fibrosis through its differntiation into pathogenic Th2 cells. This dataset includes previously published scRNA-seq data from [Min-Ji Cho et al., 2023] To investigate the tissue-specific roles of Pdcd1 in CD4⁺ T cells, bulk RNA sequencing was performed on Pdcd1^high and Pdcd1^low CD4⁺ T cells isolated from the spleen and lungs of mice. Differential gene expression analysis revealed that Pdcd1^high CD4⁺ T cells in both tissues were enriched for Th2- and fibrosis-related gene signatures. Notably, chemokine receptor expression profiles differed between spleen and lung, suggesting tissue-specific transcriptional programs associated with Pdcd1 expression.

An oncolytic virus–delivered TGFβ inhibitor overcomes the immunosuppressive tumor microenvironment

Project description:Oncolytic viruses (OV) are promising forms of immunotherapy that have demonstrated clinical benefit in difficult-to-treat cancers such as metastatic melanoma. However, their adoption in other malignancies has been limited, in part, due to poorly understood mechansims of therapeutic resistance. Here, bulk RNA-seq was performed on oncolytic vaccinia virus-sensitive and -resistant murine head and neck squamous cell carcinomas (MEERvvS and MEERvvR, respectively) to explore potential means of OV resistance. These results corroborated a potential role for TGF-beta mediated stabilization of immunosuppressive regulatory T cells in the tumor microenvironment of OV-resistant MEERvvR-bearing mice. Subsequently, treatment with oncolytic vaccinia virus engineered to expess a dominant negative TGF-β signaling inhibitor (VVtgfbi) restored sensitivity to OV-mediated cell death among MEERvvR tumors. Single-cell RNA seq performed on CD45+ immune cells isolated from tumors suggests TGF-β inhibition may also reduce the presence and activity of myeloid-derived suppressor cell populations within the tumor microenvironment.

Transcriptional profiling revealed a unique Treg signatures in SM, M or SM + M relative to control mice. Conclusion: Our study represents the first detailed analysis of splenic Treg transcriptome in mice exposure to smoke plus morphine.

Identifying genetic expression patterns as marker for sensitivity to oncolytic viruses using patient-derived pancreatic ductal adenocarcinoma organoids

Project description:Purpose: Pancreatic ductal adenocarcinoma (PDAC) is among the deadliest cancers, with a poor prognosis. Viro-immunotherapy using oncolytic viruses (OVs) represents a promising treatment for PDAC. However, PDACs from different patients respond differently to OV therapies, highlighting the need for predictive biomarkers to optimize outcomes. This study aimed to identify gene expression profiles (GEPs) that could serve as predictors for OV sensitivity. Experimental design: Patient-derived organoids (PDOs) from ten PDAC patients were assessed for sensitivity to various OVs: Newcastle Disease Virus, a Reovirus strain entering cells independently of JAM-A expression, the Edmonston vaccine strain of Measles Virus, and a gorilla-derived adenovirus. To identify GEPs with predictive value, OV sensitivity was correlated with the transcriptome of uninfected PDOs. These GEPs were analyzed in the Gene Ontology database to reveal biological processes associated with them. Results: PDOs showed heterogenic responses to OVs, with nine of ten being sensitive to at least one. Analysis of the generated GEPs using the Gene Ontology database identified intracellular aberrations as the primary determinant of OV sensitivity. Sensitivity to all OVs was associated with GEPs related to embryonic development, while varied sensitivity of the ten PDOs to four OVs was linked to cell cycle regulation, metabolism, and proliferation. Conclusions: This study revealed that OV sensitivity was associated with GEPs comprising genes involved in multiple biological processes. Further investigation into screening tumors for the identified GEPs may yield biomarkers for selecting the most effective OV for PDAC. This approach is a stepping stone toward personalized OV treatments.

The heterogeneous sensitivity of pediatric brain tumors to different oncolytic viruses is predicted by unique gene molecular profiles

Project description:Despite decades of research, the prognosis of high-grade pediatric brain tumors (PBTs) remains dismal, however, recent cases of favorable clinical responses were documented in clinical trials using oncolytic viruses (OVs). Therefore, we employ four different species of OVs: adenovirus Delta24-RGD, herpes simplex virus rQNestin34.5v1, reovirus R124 and the non-virulent Newcastle Disease Virus rNDV-F0-GFP against three entities of PBTs (high-grade gliomas, atypical teratoid/rhabdoid tumors and ependymomas) to determine their in vitro efficacy. These four OVs were screened on 14 patient-derived spheroids and the degree of oncolysis was assessed using an ATP-based assay. Subsequently, the observed viral efficacies were correlated to whole transcriptome data and Gene Ontology analysis was performed. Although no significant tumor type-specific OV efficacy was observed, whole transcriptome analysis revealed the intrinsic biological processes that associated with OV efficacy. The predictive power of those molecular profiles was further validated by screening the OV in vitro efficacy on additional PBTs. In summary, our results demonstrate OV susceptibility of multiple patient-derived PBT entities and the ability to predict in vitro responses to OVs using unique molecular profiles. Those profiles highlight the cellular intrinsic factors for potent oncolysis, that could potentially allow stratification of patients and lead to improved OV responses.

Please stand by: how oncolytic viruses impact bystander cells.

Project description:Oncolytic viruses (OVs) do more than simply infect and kill host cells. The accepted mechanism of action for OVs consists of a primary lytic phase and a subsequent antitumor and antiviral immune response. However, not all cells are subject to the direct effects of OV therapy, and it is becoming clear that OVs can also impact uninfected cells in the periphery. This review discusses the effects of OVs on uninfected neighboring cells, so-called bystander effects, and implications for OV therapies alone or in combination with other standard of care chemotherapy.
