ABSTRACT: HaCAT cells were treated with pentafactor (M5) to construct a psoriasis cell model, which was used to study the pathogenesis of psoriasis at the in vitro level. This study aimed to explore the genes activated by pentafactor (M5) treatment.

A global transcriptome analysis of human keratinocyte cell line HaCaT overexpressing the miR-21-3p.

Project description:Psoriasis is a chronic inflammatory skin disease that is mediated by a complex crosstalk between immune cells and keratinocytes (KCs). Emerging studies showed a specific psoriatic microRNAs signature where miR-21 is one of the most upregulated miRNAs. In this study, we focused our investigations on the passenger miR-21-3p strand that is poorly studied in skin and in psoriasis pathogenesis. Here, we transfected the HaCaT cells with synthetic oligoncucleotides form of miR-21-3p and evaluated the impact of the over-expression of this miRNA in these call by RNA-seq analysis. We identified a network of differentially expressed genes involved in abnormal proliferation control and immune regulatory genes implicated in the molecular pathogenesis of psoriasis in response to miR-21-3p overexpression in KCs. These results were confirmed by functional assays that validate the proliferative potential of miR-21-3p. All these findings highlight the importance of miR-21-3p, an underestimated miRNA, in psoriasis and provide novel molecular targets for therapeutic purposes.

Study of transcriptional effects of monosodium urate crystal (MSU) on HaCaT keratinocyte cell line.

Project description:Aim of the study was to characterize at a molecular level (changes in transcriptomes) the effect of monosodium urate crystal (MSU) on HaCaT keratinocyte cell line. This was adressed by using a culture model. The HaCaT cell line (human keratinocytes) was stimulated by MSU (1mg/mL) vs control for 12 hrs. By using genome-wide expression profiling, we identified deregulation of functionally relevant gene networks. HaCaT were obtained from Cell Lines Service (Eppelheim, Germany) and grown in DMEM medium (PAN biotech, Aidenbach, Germany) supplemented with 10% FBS (Life Technology, Grand Island, NY, USA), L-glutamine and non-essential amino acid. Before the treatment HaCaT cells were cultured in serum-free medium for 12hrs. HaCaT were treated with MSU (1mg/ml) vs DMEM control for 12hrs then submitted to RNA extration and gene expression profiling. Triplicate experiments were performed: HaCaT control (n=3), MSU-treated (n=3).

Livin is upregulated in keratinocytes and promotes the release of inflammatory mediators in psoriasis

Project description:Psoriasis is a common chronic inflammatory skin disease. Keratinocytes (KCs) are important effector cells that can recruit inflammatory cells by releasing inflammatory factors and chemokines to promote the inflammatory cascade in psoriasis. However, the mechanism underlying KC activation in psoriasis remains unclear. Livin is an inhibitor of apoptotic proteins and its expression can directly affect the proliferation and metastasis of tumor cells. Livin expression has been reported to be significantly increased in the lesions of patients with psoriasis; however, its specific role in KC activation has not yet been reported. The aim of this study was to investigate whether livin regulates KC activation and causes the release of inflammatory mediators. The expression levels of livin in patients with psoriasis, an imiquimod (IMQ) mouse model, and M5-treated HaCaT cells were determined via immunofluorescence staining, reverse transcription-quantitative polymerase chain reaction, enzyme-linked immunosorbent assay (ELISA), and western blotting. We constructed livin knockdown (Knockdown-HaCaT) and negative control (NC-HaCaT) cells using human immunodeficiency virus-1-based lentiviral vectors to study the function of livin in KCs via RNA-sequencing and proteomics analysis. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were performed. Moreover, the effect of livin expression on the release of inflammatory mediators in KCs was verified using ELISA.

Extremely low frequency electromagnetic fields (ELF-EMFs) effect on the genetic expression profile of a human keratinocyte line (HaCaT)

Project description:Clinical evidence has shown that electromagnetic fields produce a benefical therapeutic result for wounds, however little is still known about their exact mechanism of action. Moreover, clinical results concerning skin tissue restoration are still debated. In the present study, we carried out gene expression profiling of a human keratinocyte line (HaCaT) submitted to 1 hour of ELF-EMF (frequency 50Hz, intensity 1 mT) in order to identify up- and downregulated genes by this stimulus, and to verify the presence of specific molecular pathways activation. Most of the genes modulated were involved in mechanisms such as protein synthesis. In particular, these genes are related to Mammalian target of Rapamycin (mTOR), which has been identified as a kinase with a pivotal role in cellular proliferation and survival in mammals. In this study, we analyzed the expression profiles of 3 biological replicates of HaCaT cells exposed to ELF-EMF (frequency 50Hz, intensity 1 mT) for 1 hour. All HaCaT cells exposed to ELF-EMF RNAs were hybridized against control sham-exposed HaCaT cells RNAs. Each biological sample was repeated with a technical replicate (extraction-labeling) and a dye-swap experiment.
