Transcriptomic analysis of ovarian cancer cells after drug administration and tRNA transfection at 10 and 24 hours

ABSTRACT: SLFN11 sensitizes cancer cells to DNA-damaging agents (DDAs) by cleaving and reducing tRNALeu(TAA). In this study, we investigated how the reduction of tRNALeu(TAA) contributes to cell death under Camptothecin (CPT) treatment. RNA-seq and proteome analyses were performed to assess the global effects of tRNALeu(TAA) transfection. The results suggest that tRNALeu(TAA) influences the expression of proteins essential for proteostasis, particularly those involved in ubiquitin-dependent proteolysis, at the protein level rather than the mRNA level. These findings indicate that SLFN11 plays a crucial role in proteostasis by regulating tRNAs, and thus determines cell fate under DDA treatment.
