Brassinosteroids (BRs) play crucial roles during hypocotyl establishment both in the light and darkness. Arabidopsis brassinosteroid insensitive1 (bri1-5) is a partial loss-of-function mutant, and the short-hypocotyl phenotype of bri1-5 is greatly suppressed by a long-hypocotyl mutant phytochrome B (pbyB-77) at the double mutant for the two in the light conditions to display additive phenotypes of both. To identify the genes responsible for this genetic interaction, we employed microarray analysis with the RNAs prepared from wild type, bri1-5, phyB-77, and bri1-5 phyB-77 double mutant. Microarray analysis supported the notion that PHYB acts upstream of BRs.

Project description:Light is a major determinant of plant growth and survival. NONEXPRESSER OF PATHOGENESIS-RELATED GENES 1 (NPR1) acts as a receptor for salicylic acid (SA) and serves as the key regulator of SA-mediated immune responses. However, the mechanisms by which plants integrate light and SA signals in response to environmental changes, as well as the role of NPR1 in regulating plant photomorphogenesis, remain poorly understood. This study shows that SA promotes plant photomorphogenesis by regulating PHYTOCHROME INTERACTING FACTOR 4 (PIF4). Specifically, NPR1 promotes photomorphogenesis under blue light by facilitating the degradation of PIF4 through light-induced polyubiquitination. NPR1 acts as a substrate adaptor for the CULLIN3-based E3 ligase, which ubiquitinates PIF4 at Lys129, Lys252, and Lys428, and leading to PIF4 degradation via the 26S proteasome pathway. Genetically, PIF4 is epistatic to NPR1 in the regulation of blue light-–induced photomorphogenesis, suggesting it acts downstream of NPR1. Furthermore, cryptochromes mediate the polyubiquitination of PIF4 by NPR1 in response to blue light by promoting the interaction and ubiquitination between NPR1 and PIF4. Transcriptome analysis revealed that, under blue light, NPR1 and PIF4 coordinately regulate numerous downstream genes related to light and auxin signaling pathways. Overall, these findings unveil a role for NPR1 in photomorphogenesis, highlighting a mechanism for post-translational regulation of PIF4 in response to blue light. This mechanism plays a pivotal role in the fine-tuning of plant development, enabling plants to adapt to complex environmental changes.

Project description:The developmental switch from skotomorphogenesis to photomorphogenesis is critical for the survival and growth of plants, but its regulatory mechanism remains unclear. Here, we report that the steroid hormone brassinosteroids (BRs) play crucial roles in the transition from skotomorphogenesis to photomorphogenesis by regulating chlorophyll biosynthesis to promote the greening of etiolated seedlings upon light exposure. Seedlings of BR-deficient mutant det2-1 accumulated excess protochlorophyllide when grown in darkness, resulting in photo-oxidative damage upon exposure to light. Conversely, the gain-of-function mutant bzr1-1D suppressed the protochlorophyllide-accumulated phenotype of det2-1, thereby promoting greening of etiolated seedlings. Genetic analysis indicated that phytochrome-interacting factors (PIFs) were required for BZR1-promoted seedlings greening. Furthermore, we revealed that the GROWTH REGULATING FACTOR 7 (GRF7) and GRF8 were induced by BZR1 and PIF4 to repress the chlorophyll biosynthesis and promote seedling greening. Suppression the functions of GRFs by overexpressing microRNA396a (miR396a) caused the high-accumulated photochlorophyllide in darkness and more serious photobleach upon light exposure. Additionally, BZR1, PIF4 and GRF7 interact with each other and precisely regulate the expression of chlorophyll biosynthetic genes. Our findings revealed an essential role of brassinosteroid in promoting seedling development and survival during the critical initial emergence of seedlings from subterranean darkness to sunlight.
