Transcriptome-Wide M5C Methylation Profiling in NSUN6-Knockdown HepG2 Cells

ABSTRACT: This study investigates the role of NSUN6 in RNA 5-methylcytosine (M5C) methylation in HepG2 cells. Stable NSUN6 knockdown cell lines were generated using shRNA-mediated RNA interference, validated by qRT-PCR and Western blot. RNA was extracted from knockdown and control cells, treated with bisulfite to convert unmethylated cytosines, and sequenced. Sequencing data were aligned to the human genome (GRCh38) to identify M5C methylation sites. By comparing methylation levels between NSUN6 knockdown and control cells, we aim to map NSUN6-dependent M5C sites and elucidate their roles in regulating gene expression and cellular function. This dataset provides valuable insights into the epigenetic regulation of RNA modifications and the specific contributions of NSUN6 in human cells, with potential implications for understanding disease mechanisms.
