Transcrpitomic analysis of Oral lichen planus and oral lichenoid lesions

ABSTRACT: This dataset contains bulk RNA sequencing profiles of human buccal mucosal tissues collected from 42 individuals, including 10 healthy controls, 16 patients with oral lichen planus (OLP), and 16 patients with oral lichenoid lesions (OLL). The objective of this study was to investigate the immune heterogeneity underlying OLP and OLL by comparing transcriptomic differences among the three groups. All tissue samples were obtained from the buccal mucosa, and RNA-seq was performed to identify immune-related gene expression patterns and molecular signatures. This dataset provides a valuable resource for exploring the immunopathogenesis of OLP and OLL.

METATAXONOMIC AND METAPROTEOMIC PROFILING OF THE ORAL MICROBIOME IN ORAL LICHEN PLANUS

Project description:Background: A growing body of evidence demonstrates a different bacterial composition in the oral cavity of patients with oral lichen planus (OLP). Patients and methods: Buccal swab samples were collected from affected and non-affected sites of six patients with reticular OLP and the healthy oral mucosa of six control subjects. 16S rRNA MiSeq sequencing and mass spectrometry-based proteomics and were utilised to identify the metataxonomic and metaproteomic profiles of the oral microbiome in both groups. Results: The most abundant species in the three subgroups were Streptococcus oralis and Pseudomonas aeruginosa, accounting for up to 70% of the total population. A Canonical Correspondence Analysis showed differential clustering of samples from the healthy and OLP groups. Three species (Veillonella parvula, Actinomyces sp, and Lactococcus lactis) were significantly over-represented in the control group and one (Granulicatella elegans) in patients with OLP. The metaproteomic data revealed that several G. haemolysans-belonging peptidases and other proteins with inflammatory and virulence potential were found present in OLP lesions only. Conclusion: Our data suggest that several bacterial species and peptides are associated with OLP. Future studies with larger cohorts should be conducted to determine their role in the aetiology of OLP and evaluate their potential as disease biomarkers.

Project description:Oral lichen planus (OLP) is a poorly understood chronic inflammatory mucocutaneous disease and is associated with many systemic diseases, which may lead to the erosion of oral mucosa and bring physical and psychological disorders to patients. However, the pathogenesis and immune microenvironment of OLP with different clinical subtypes are unclear. therefore, this study established a clinical cohort of OLP with 1-year follow-up and obtained the clinical information and bulk RNA-seq files to explore the immune microenvironment and molecular mechanism of oral lichen planus with different clinical subtypes. Based on the erosion of oral mucosa in the biopsy, the participants of OLP were divided into two groups, non-erosive OLP (NEOLP) and erosive OLP (EOLP). According to whether erosion occurred in oral mucosa more than three times and a remission period of less than 3 months within a one-year follow-up, OLP was divided into two groups, recalcitrant erosive OLP (REOLP) and stable OLP (SOLP).

Transcriptome sequencing of primary human oral keratinocytes co-cultured with Prevotella melaninogenica

Project description:Oral lichen planus (OLP) is a common oral mucosal disease, of which the etiology and pathogenesis are still unclear. Microorganisms may play an essential role in the pathogenesis of OLP. Previous studies of our group found that the composition ratio of Prevotella melaninogenica (Pm) on the buccal mucosa surface of OLP patients increased significantly. In addition, Pm could invade the epithelium of OLP. As the first physical defense of the oral mucosa, oral keratinocytes may interact directly with Pm in OLP. Therefore, this study aimed to explore the impact of Pm on primary human oral keratinocytes' transcriptome.

Project description:In this study, we compared microRNA (miRNA) profiles of salivary exosomes of patients with oral lichen planus with those of healthy controls. Saliva samples from 16 patients with oral lichen planus and 8 healthy controls were divided into 2 sets and were examined by performing miRNA microarray analysis. Examination of 8 oral lichen planus patients and 4 healthy controls. Each patient and control represent pooled RNAs from salivary exosomes of 8 patients and 4 healthy controls, respectively. Please note that each set (i.e. set1 and set2) was analysed independently.
