Effects of attenuating astrocytic Ca2+ activity in the medial prefrontal cortex with iβARK on transcriptional profiles in wild-type mice.

ABSTRACT: We injected adeno-associated viruses (AAV2/5) carrying an inhibitory peptide derived from β-adrenergic receptor kinase 1 (iβARK), tagged with mCherry for visualization, into the medial prefrontal cortex (mPFC) of wild-type mice. This approach selectively attenuated Gq-coupled GPCR-induced Ca²⁺ activity in astrocytes. Control mice received AAV2/5 expressing mCherry alone. Subsequently, we performed RNA sequencing on mPFC tissue samples to compare transcriptomic profiles and investigate the potential functional consequences of perturbing Gq-GPCR signaling.
