Single-nuclus RNA sequencing of stem cell-derived human trophoblast organoids cultured under simulated microgravity

ABSTRACT: Human placental development requires precise cellular differentiation, yet mechanistic understanding remains limited. Here, we perform single-nucleus RNA sequencing of day 11 human trophoblast organoids cultured under simulated microgravity to characterize cellular diversity and lineage specification, revealing distinct populations including cytotrophoblasts, syncytiotrophoblasts, and extravillous trophoblasts.

Single-nucleus RNA-seq analysis of extravillous trophoblast differentiation from human trophoblast organoids cultured under simulated microgravity

Project description:To investigate extravillous trophoblast (EVT) differentiation mechanisms, we generated human trophoblast organoids (hTOr) from human trophoblast stem cells (hTSCs) and analyzed them by single-nucleus RNA sequencing. Organoids were initially cultured under simulated microgravity using a ClinoStar bioreactor for 11 days to establish proper trophoblast architecture, then embedded in Matrigel and cultured in EVT differentiation medium for 14 days. This study examines the cellular composition and differentiation trajectories during EVT specification, providing insights into human placental development and trophoblast lineage decisions. Two independent hTSC lines (CT27 and CT29) were used as biological replicates to ensure reproducibility and capture potential line-specific variations in EVT differentiation capacity.asts.

Single-nucleus RNA sequencing of human trophoblast-mesoderm assembloids

Project description:Human placental and extraembryonic development involves complex interactions between trophoblast and mesoderm lineages. Here, we performed single-nucleus RNA sequencing of day 11 and day 23 trophoblast-mesoderm assembloids generated by co-culturing human trophoblast stem cells (hTSCs) with extraembryonic mesoderm-primed (ExM) cells under rotational culture conditions. Our analysis reveals distinct cellular populations including cytotrophoblasts, syncytiotrophoblasts, extravillous trophoblasts, and mesodermal derivatives, providing insights into lineage specification and cell-cell interactions during early human development.

Effects of Simulated Microgravity on Colorectal Cancer Organoids Growth and Drug Response

Project description:Cellular and molecular dynamics of human cells are constantly affected by gravity. Alteration of the gravitational force disturbs the cellular equilibrium, which might modify physiological and molecular characteristics. Nevertheless, biological responses of cancer cells to reduced gravitational force remains obscure. Here, we aimed to comprehend not only transcriptomic patterns but drug responses of colorectal cancer (CRC) under simulated microgravity. We established four organoids directly from CRC patients, and organoids cultured in 3D clinostat were subjected to genome wide expression profiling and drug library screening. Our observations revealed changes in cell morphology and an increase in cell viability under simulated microgravity compared to their static controls. Transcriptomic analysis highlighted a significant dysregulation in the TBC1D3 family of genes. The upregulation of cell proliferation observed under simulated microgravity conditions was further supported by enriched cell cycle processes, as evidenced by the functional clustering of mRNA expressions using cancer hallmark and gene ontology terms. Our drug screening results indicated an enhanced response rate to 5-FU under conditions of simulated microgravity, suggesting potential implications for cancer treatment strategies in simulated microgravity.

Our results indicate that the retinoic acid inducible gene (RIG)-I-like receptor (RLR) and Toll-like receptor (TLR) signal pathways, which are both involved in the type-I interferon induction, are significantly inhibited by simulated microgravity effects.

Limnospira indica PCC8005 in simulated microgravity

Project description:Regenerative life support systems for space crews recycle organic and inorganic waste into water, food and oxygen using different organisms. For instance, the European Space Agency's MELiSSA uses Limnospira indica PCC8005 for air revitalisation and food production. Before use in space, the components' compatibility with reduced gravity must be tested. This innovative study introduces a novel ground analog designed specifically for microgravity experiments involving cyanobacteria, employing a cutting-edge random positioning machine (RPM). Limnospira indica PCC8005 was shown to grow slower under simulated microgravity and whole proteome analysis revealed a downregulation of e.g. ribosomal proteins, glutamine synthase and nitrate uptake transporters while an upregulation was found for gas vesicle proteins, carboxysome proteins and phycobiliproteins. All together our results suggested that L. indica experienced carbon limitation when cultivated in simulated microgravity conditions.

Matrix directs trophoblast differentiation in a bioprinted organoid model of early placental development

Project description:Trophoblast organoids offer a unique opportunity to study mechanisms orchestrating placental growth and development during pregnancy. However, many organoid cultures rely on extracellular matrix reagents that are highly variable and unable to be tuned to reflect in vivo tissues. Here we describe the first bioprinted placental organoid model, generated using first trimester trophoblast cell line, ACH-3P, and a synthetic polyethylene glycol matrix. Bioprinted organoids were compared comprehensively to classic Matrigel-embedding using functional assays, immunofluorescence microscopy, transcriptomic and proteomic analyses. Organoids differentiated spontaneously from cytotrophoblasts into two major subtypes: extravillous trophoblasts (EVTs) and syncytiotrophoblasts (STBs), with bioprinted organoids driven towards EVT differentiation. Bioprinted organoids were exposed to inflammation and treated with aspirin or metformin to assess their effects on trophoblast organoid formation and viability. Further, we reversed the inside-out architecture of ACH-3P organoids by suspension culture. Organoid suspension caused STBs to form a syndecan-1+ outer layer on the periphery of organoids, reflecting placental tissue. Here, we present an alternative trophoblast organoid model with further tuning potential to reflect the placental microenvironment in physiological and pathological pregnancies.
