Project description:Plant memory is a tool for plants that they can use to increase their fitness and cope with adverse environmental stresses. In this study, mRNA-seq., WGBS and RT-qPCR methods were applied for evaluating formation and maintenance of somatic transcriptional memory after treatment with ultrasound and drought stimuli in tomatoes. In addition, effects of repeated stimuli, as well as association-forming ability of plants were studied when they were trained previously with combined stimuli. 48 h after exposure to the two stimuli applied alone or in combination, significantly altered gene transcription and DNA methylation were revealed but there was no correlation between these changes. Using four selected target genes, we have proven that plants memorized stimuli for 5 to 10 days, in a gene-, and stimulus-dependent way. The repeated application of the stimuli caused altered behavior, such as habituation, sustained induction or modified re-induction, depending on the genes and physiological ages of plants. Plants were able to use one conditioned stimulus as a predictor of the other, unconditioned one, after conditioning in the case of 3 out of 4 target genes, and use their transcriptional memory associatively.

Project description:In a scenario of global climate change, water scarcity is a major threat for agriculture, severely limiting crop yields. Therefore, alternatives are urgently needed for improving plant adaptation to drought stress. Among them, gene expression reprogramming by microRNAs (miRNAs) might offer a biotechnologically sound strategy. Drought-responsive miRNAs have been reported in many plant species, and some of them are known to participate in complex regulatory networks via their regulation of transcription factors involved in water stress signaling. We explored the role of miR159 in the response of Solanum lycopersicum Mill. plants to drought stress by analyzing the expression of sly-miR159 and its target SlMYB transcription factor genes in tomato plants of cv. Ailsa Craig grown in deprived water conditions or in response to mechanical damage caused by the Colorado potato beetle, a devastating insect pest of Solanaceae plants. Results showed that sly-miR159 regulatory function in the tomato plants response to distinct stresses might be mediated by differential stress-specific MYB transcription factor targeting. sly-miR159 targeting of SlMYB33 transcription factor transcript correlated with accumulation of the osmoprotective compounds proline and putrescine, which promote drought tolerance. This highlights the potential role of sly-miR159 in tomato plants' adaptation to water deficit conditions.

Project description:Grafting is widely used as a method to increase stress tolerance in good fruiting lines of Solanaceae plants. However, little is known about how grafting, affects epigenetic modifications and leads to stress tolerance, especially within the same line. Here, we studied the effects of self-grafting in tomato plants on histone and DNA modifications and changes in gene expression related to drought stress. We found that at the three-leaf stage, 1 week after self-grafting, histone H3 K4 trimethylation and K27 trimethylation changes were observed in more than 500 genes each, and DNA methylation changes in more than 5,000 gene regions at the shoot apex compared to the non-grafted control. In addition, two weeks after the epigenomic changes, global expression changes continued to be observed at the shoot apex in several genes related to the metabolic process of nitrogen compounds, responses to stimulus, chromosome organization, cell cycle-related genes, and regulation of hormone levels. Finally, these grafted seedlings acquired remarkable drought tolerance, suggesting that epigenomic modifications during the wound-healing process mitigate stress tolerance in tomato plants.

Project description:Global climate change exacerbates abiotic stresses, as drought, heat, and salt stresses are anticipated to increase significantly in the coming years. Plants coexist with a diverse range of microorganisms. Multiple inter-organismic relationships are known to confer benefits to plants, including growth promotion and enhanced tolerance to abiotic stresses. In this study, we investigated the mutualistic interactions between three fungal endophytes originally isolated from distinct arid environments and an agronomically relevant crop, Solanum lycopersicum. We demonstrated a significant increase in shoot biomass under drought conditions in co-cultivation with Penicillium chrysogenum isolated from Antarctica, Penicillium minioluteum isolated from the Atacama Desert, Chile, and Serendipita indica isolated from the Thar Desert, India. To elucidate plant gene modules commonly induced by the different endophytes that could explain the observed drought tolerance effect in tomato, a comprehensive transcriptomics analysis was conducted. This analysis led to the identification of a shared gene module in the fungus-infected tomato plants. Within this module, gene network analysis enabled us to identify genes related to abscisic acid (ABA) signaling, ABA transport, auxin signaling, ion homeostasis, proline biosynthesis, and jasmonic acid signaling, providing insights into the molecular basis of drought tolerance commonly mediated by fungal endophytes. Our findings highlight a conserved response in the mutualistic interactions between endophytic fungi isolated from unrelated environments and tomato roots, resulting in improved shoot biomass production under drought stress.

Project description:Although glutathione S-transferases (GST, EC 2.5.1.18) are involved in response to abiotic stress, limited information is available regarding gene function in tomato. In this study, a GST gene from tomato, designated LeGSTU2, was cloned and functionally characterized. Expression profile analysis results showed that it was expressed in roots and flowers, and the transcription was induced by salt, osmotic, and heat stress. The gene was then introduced to Arabidopsis by Agrobacterium tumefaciens-mediated transformation. Transgenic Arabidopsis plants were normal in terms of growth and maturity compared with wild-type plants. Transgenic plants also showed an enhanced resistance to salt and osmotic stress induced by NaCl and mannitol. The increased tolerance of transgenic plants was correlated with the changes in proline, malondialdehyde and antioxidative emzymes activities. Our results indicated that the gene from tomato plays a positive role in improving tolerance to salinity and drought stresses in Arabidopsis.

Project description:Drought is a key environmental factor that restricts crop growth and productivity. Plant responses to water-deficit stress at the whole plant level are mediated by stress-response gene expression through the action of transcription factors (TF). The NAC (NAM/ATAF/CUC) transcription factor family has been well documented in its role in improving plant abiotic stress tolerance. In the present study we evaluated the effects of overexpression of SlNAC2 TF on the photosynthetic machinery, relative water content (RWC), reactive oxygen species, antioxidants and proline levels in tobacco plants exposed to a water-deficit treatment. Shoot growth and seed formation were also evaluated before, during and following water-deficit to determine any morphological consequences of transgene expression. The transgenic plants maintained higher RWC and chlorophyll levels over 21 days after withholding water and stomatal conductance until the 16th day of water-deficit. Overexpression of SlNAC2 in tobacco increased proline levels, improved seed setting and delayed leaf senescence of the transgenic plants. Reactive oxygen species accumulated at lower levels in the dehydrated transgenic plants but no significant difference in superoxide dismutase and catalase content were seen between the genotypes. The conversion of glutathione to oxidized glutathione was significantly higher in the transgenic plants, supported by increased glutathione reductase transcript levels. Our results indicate that overexpression of SlNAC2 in tobacco improved survival during and recovery from water-deficit stress, without an associated biomass penalty under irrigation.Supplementary informationThe online version contains supplementary material available at 10.1007/s12298-021-00996-2.
