ABSTRACT: Conventional dendritic cells (cDCs) has been shown to mediate immune checkpoint inhibitor responses in cancer patients. We used single nucleus ATAC sequencing (snATAC-seq) to analyze the epigenome of heterogeneous cDCs obtained from 14 human metastatic melanoma samples.

Conventional dendritic cells of lymphoid origin with a preferential distribution to barrier tissues [cDCs_scRNA-seq]

Project description:Conventional dendritic cells (cDCs) regulate adaptive immunity. Although cDCs emerged with the evolution of adaptive immunity, it is considered that most cDCs are of myeloid origin (M-cDCs). Using a lymphoid progenitor-tracking system, we identify novel cDCs of lymphoid origin (L-cDCs) in a steady state. Notably, L-cDC2s are predominantly distributed in barrier tissues such as the lungs and skin. Single-cell RNA-sequencing analysis revealed that the distribution and gene expression patterns of L-cDC clusters are very similar to M-cDC clusters, except for the enrichment of lymphocyte-signature genes in L-cDCs. Importantly, we identified lymphoid-derived pre-DCs and lymphocyte-primed cDC precursors as sources of L-cDCs. Compared with M-cDC2s, L-cDC2s weakly primed T cells under low-dose antigen stimulation and preferentially promoted Th2 differentiation under sufficient antigenic stimulation. These results suggest the developmental diversity of cDCs and imply the contribution of L-cDCs to tolerance and hyper-responses to Th2-related antigens in barrier tissues.

Klf4 expression in conventional dendritic cells is required for T helper 2 cell responses

Project description:Subset-specific and progenitor gene expression analysis of Klf4+/+ and Klf4-/- DCs. The two major lineages of classical dendritic cells (cDCs) express and require either IRF8 or IRF4 transcription factors for their development and function. IRF8-dependent cDCs promote anti-viral and T-helper 1 (Th1) cell responses, whereas IRF4-expressing cDCs have been implicated in controlling both Th2 and Th17 cell responses. Here, we have provided evidence that Kruppel-like factor 4 (Klf4) is required in IRF4-expressing cDCs to promote Th2 but not Th17 cell responses in vivo. Conditional Klf4 deletion within cDCs impaired Th2 cell responses during Schistosoma mansoni infection, Schistosoma egg antigen (SEA) immunization, and house dust mite challenge (HDM), without affecting cytotoxic T lymphocyte (CTL), Th1 and Th17 cell responses to herpes simplex virus, Toxoplasma gondii and Citrobacter rodentium infections. Further, Klf4 deletion reduced IRF4 expression in pre-cDCs and resulted in selective loss of IRF4-expressing cDCs subsets in several tissues. These results indicate that Klf4 guides a transcriptional program promoting IRF4-expressing cDCs heterogeneity. Bone marrow progenitors and skin draining LN subsets were harvested from 4 Klf4fl/fl cre negative or Vav1-icre mice and were sorted to >95% purity on the FACS Aria 3.

Transcription profiling of human plasmacytoid dentritic cells co-cultured with activated conventional dendritic cells

Project description:Two well-characterized blood dendritic cell populations, conventional (cDC) and plasmacytoid (pDC), exhibit multiple phenotypic, migratory and functional differences that suggest specialized and may be complementary and coordinated functions. To study this possible coordination, cDCs and pDCs from healthy blood donors were sorted and cDCs were stimulated either with LPS or R848 whereas pDCs were CFSE-labelled and maintained in IL3. Then, CFSE labelled-pDCs and stimulated-cDCs were mixed and co-cultured. Following this "conditioning", CFSE-pDCs were sorted again and further analyzed. Conditioned pDCs showed moderate phenotypic maturation and acquired allostimulatory capacity. Microarray and RT-PCR analyses showed the induction of different genes including chemokines and proinflammatory cytokines that were dependent on the stimulation received from the "conditioner" cDC. Additionally, the differential pattern of conditioning was confirmed by protein secretion analyses with the production of specific chemokines and cytokines by conditioned pDCs. Importantly, conditioning of pDCs by activated cDCs required cell-cell contact.
