Effect of overexpression of VGF on gene expression of A549 and H1299 NSCLC cells

ABSTRACT: This study investigates the transcriptional changes induced by VGF overexpression in A549 and H1299 NSCLC cell lines. RNA-seq was performed on cells transfected with a VGF lentivirus or an empty vector control. The goal is to identify dysregulated genes and pathways mediated by VGF overexpression in lung cancer cells.

Project description:Non-small cell lung cancer (NSCLC) patients are prone to drug resistance during chemotherapy. Therefore, in order to compare the changes in the gene expression profiles of NSCLC cells before and after drug resistance, we constructed cisplatin-resistant cells (NCI-H1299/CDDP), and compared the gene expression profiles with those of the parental NCI-H1299 cells. We used microarrays to study in detail the global gene expression changes before and after drug resistance in NSCLC cells and identified genes that were up- or down-regulated during this process.

MicroRNA expression data from H1299 cells

Project description:We explored the biological effects of AGO2 downregulation on the expression of miRNAs in human NSCLC cells. Using AGO2 siRNA as a positive control, we overexpressed miR-138 in H1299 cells to observe changes in the miRNA profile using an miRNA microarray assay. Total RNAs were extracted from H1299 cells transfected with NC, AGO2 siRNA or miR-138 for 48h. Their microRNAs expression profiles were investigated using LCSciences Human miRNA Array platform (LCSciences, Houston, TX; Sanger mirBase Release 19.0).
