Human IgG1 (B-cell receptor) Fc peptide glycan analysis, LC-MS

Project description:Since few data are available on glycans expressed by human BCRs, we aimed to analyze the Fc glycans of membrane-bound IgG after BCR capture and tryptic digestion by LC-MS. For this purpose, human B-cell lines from Burkitt Lymphoma (Ramos) expressing IgG1-BCRs in the presence or absence of Fc glycans (FcG) were analyzed. The B-cell lines were generated by transduction of the BCR-negative MDL-AID KO cell line with N(297) or mutant Q(297) IgG-BCRs. The BCR sequences used were derived from single-cell sorted human B cells isolated from patients with the autoimmune disease rheumatoid arthritis. Two BCRs were directed towards citrullinated antigens (2G9 and 3F3), while a third BCR was directed against tetanus toxoid (D2). In addition, we analyzed the Fc glycan profile of IgG secreted (sIgG) by human Ramos B cells for comparison.

Investigation of human peripheral T cells responsive to SARS-CoV-2 antigens

Project description:To elucidate the T cell epitopes of SARS-CoV-2, we stimulated human PBMCs from healthy donors and convalescent COVID-19 patients with various SARS-CoV-2 antigens, sorted the activated T cells and performed sc-RNA and -TCR sequencing. We obtained thousands of T cell clonotypes that responded to SARS-CoV-2 antigens, and identified the epitopes and restricting HLAs of several clonotypes that were significantly expanded in the COVID-19 patients.

Vaccine based on immune evasion mutations integrated RBD protein have broad protection against SARS-CoV-2 [BCR-seq]

Project description:Variants of SARS-CoV-2 continue to emerge and evade immunity, resulting in breakthrough infections in vaccinated populations. Continued vaccination with vaccines based on the antigens of newly emerged variants does not necessarily result in long-lasting protection. Thus, there is an urgent need for the development of vaccines with a broad spectrum of protective effects. In this study, we selected hotspot mutations in the receptor binding domain (RBD) based on immune escape properties and integrated them into the original RBD protein to obtain a complexed RBD protein (cRBD). We designed a total of three cRBD (cRBD1-3) and thoroughly evaluated their immunological properties. Compared with the BA.1 RBD protein, the cRBDs induced higher levels and broader spectrum of neutralizing antibodies, with cRBD3 being the best performer. In vivo protective capacity of cRBDs was further validated in Balb/c mice attacked by live virus. In order to investigate the reason for the broad protective efficacy of cRBD, whole blood from mice that had completed the immunization process was subjected to BCR sequencing. BCR transcriptome analysis found differences in the use of VJ pairs in IGH among the groups.

Adaptive immune responses to SARS-CoV-2 persist in the pharyngeal lymphoid tissue of children

Project description:SARS-CoV-2 infection triggers adaptive immune responses from both T and B cells. However, most studies focus on peripheral blood, which may not fully reflect immune responses in lymphoid tissues at the site of infection. To evaluate both local and systemic adaptive immune responses to SARS-CoV-2, we collected peripheral blood, tonsils, and adenoids from 110 children undergoing tonsillectomy/adenoidectomy during the COVID-19 pandemic. In order to investigate the adaptive immune response locally, we measured the neutralizing antibodies against to multiple SARS-CoV-2 virus variants and performed high dimensional flow cytometry and CITEseq including TCR and BCR analysis for deeper characterization of the immune response to SARS-CoV-2. In our CITE-seq analyses, we sorted SARS-CoV-2 S1 (spike 1) binding (Spike1+) and non-binding (Spike1-) CD19+ B cells from tonsils, adenoids, and PBMCs from two subjects with a history of COVID-19 and one uninfected control. To assess T cells at the single cell level, we sorted CD95+ CD4+ and CD95+ CD8+ T cells, identified expanded T cell clonotypes, and then compared the CDR3 sequences to those previously reported to recognize SARS-CoV-2 antigens. Our results provide evidence for persistent tissue-specific immunity to SARS-CoV-2 in the upper respiratory tract of children weeks to months after acute infection.
