Heterogeneity in B- and epithelial-cell senescence in the pathogenesis of COPD/emphysema [scRNA-seq]


ABSTRACT: Increased cellular senescence leading to premature lung aging is a key mechanism for chronic inflammation and airway remodeling in emphysematous-COPD. However, the cell-specific changes resulting in ‘senescent microenvironment’ in COPD remains elusive. To bridge this gap, we used single-cell multiomics approach to identify the differentially regulated cell types, genes/pathways and spatial organization of senescence phenotype in smoke-induced emphysema in humans. Dimensionality reduction and clustering of 121,885 cells in lung tissues from emphysematous COPD patients and healthy controls (n = 6/ group) with and without a smoking history revealed a significant enrichment (p<0.01) in the senescence associated gene sets in B-lymphocytes and multi-ciliated non-nasal (MNN) epithelial cell clusters from COPD patients. Senescent B cells were found to have dysfunctional development and maturation as denoted by alterations in expression of targets including BMP6, CCNB2, MCM4, PTEN and DDIT4 in diseased state. The senescence phenotype in MNN cells was closely linked to increased expression of senescence effector genes (p16 and p21) through both single cell and spatial transcriptomic approaches. Outcomes from single-cell techniques were validated by conducting cell intervention studies using small airway epithelial cells (SAEC; submerged) and diseased-human bronchial epithelial (D-HBE; ALI) cells in vitro. We observed increased level of p16, but not p21, in cultured SAEC cells from COPD donors. The disease-induced upregulation of p16 was attenuated upon treatment with various senolytic interventions in SAECs, but not D-HBEs. Overall, our study defines the senescence landscape of various cell types in COPD/emphysema and identified potential genes/pathways that could be targeted for interventions in future.
