Single-nucleus RNA-seq analysis of YAP5SA-induced human trophoblast organoids cultured under simulated microgravity

ABSTRACT: To investigate the role of YAP activation in human trophoblast differentiation, we generated doxycycline-inducible YAP5SA (constitutively active YAP) expressing human trophoblast stem cells and analyzed them by single-nucleus RNA sequencing. Control and YAP5SA-induced trophoblast organoids were cultured under rotational conditions using simulated microgravity for 15 days. YAP activation resulted in substantial reduction of syncytiotrophoblast population and enhanced expression of cytotrophoblast stemness-associated genes, demonstrating YAP's role as a biomechanical integrator orchestrating trophoblast lineage allocation.
