HNRNPH controls alternative splicing cooperatively on a transcriptome-wide scale [in vitro iCLIP]


ABSTRACT: The in vitro binding potential of HNRNPH1 to RNA is not available. Hence, we performed an high-throughput in vitro binding assay for recombinant MBP-HNRNPH1 using an RNA oligo library consisting of 2000 individual sequences. We used this in vitro binding assay to map binding of recombinant HNRNPH1 protein to RNA in individual nucleotide resolution. To understand the impact of RNA G-quadruplex structures for HNRNPH1 binding, we compared the binding of HNRNPH1 to GTP RNA and 7-deaza-GTP RNA.
