Transcriptome analysis of cisplatin-treated KGN human ovarian granulosa cells

ABSTRACT: Cisplatin is a major chemotherapeutic agent in cancer treatment. However, studies have shown that chemotherapy can induce aging in the body, leading to an increased risk of Premature Ovarian Insufficiency in women. Therefore, this study conducted transcriptome sequencing analysis on KGN cells after cisplatin treatment.

Mitochondrial dynamics in ovarian granulosa cells during follicular growth and in human KGN cells

Project description:In the ovary, proliferation and differentiation of granulosa cells (GCs) drive the growth of follicles. This is, in part, dependent on gonadotropins. Our immunohistochemical studies provided hints of mitochondrial biogenesis and intracellular redistribution in GCs of growing follicles. A cellular model, human KGN cells (granulosa cell tumor cells, derived from growing follicles) was used to study aspects of mitochondrial dynamics. To elevate cAMP and thereby mimic gonadotropin actions, forskolin (FSK) was used, which increased KGN cell size and mitochondrial DNA within 24 h. MitoTracker experiments and ultrastructural 3D reconstruction revealed that FSK treatment induced the formation of elaborate mitochondrial networks. H89, a protein kinase A (PKA) inhibitor, reduced network formation. A proteomic analysis indicated that FSK among others elevated levels of regulators of the cytoskeleton and the steroidogenic enzyme CYP11A1, located in mitochondria, was more than 3-fold increased, implying that cAMP/PKA-associated structural changes go in parallel with the acquisition of steroidogenic competence of mitochondria in KGN cells. In summary, in situ observations showed increases in mitochondria and suggested intracellular trafficking in GCs during follicular growth and indicated that they may partially be under the control of gonadotropins/cAMP. In line with this, elevation of cAMP in KGN profoundly affected mitochondria dynamics in a PKA-dependent manner and implicated cytoskeletal changes.

Establishing the Oocyte Proteome at the Primordial Follicle Stage and Cisplatin-induced Shifts through in situ APEX2-driven Biotinylation

Project description:The number and quality of primordial follicles play a decisive role in female fertility. The high sensitivity of oocytes in primordial follicles to chemotherapeutics makes antitumor therapy extremely prone to premature ovarian insufficiency and infertility; however, the specific mechanism remains unclear. A systematic proteomic profile of oocytes in primordial follicles is urgently needed to support basic and clinical research, which has been hindered by the rarity of oocyte samples and the technical challenges associated with isolating oocytes from primordial follicles. In this study, utilizing in vivo protein labeling by engineered ascorbate peroxidase (APEX) fluorescent mice, we identified 641 proteins and 2020 gene transcripts in primordial follicular oocytes, from which the abundance of several proteins involved in processes associated with DNA damage repair and histone modification changed after ovary exposure to the chemotherapeutic cisplatin. Notably, a histone modifier, EZH2, was induced by cisplatin in oocytes, and the simultaneous application of its inhibitor, GSK126, partially relieved cisplatin-induced oocyte developmental defects. Our study provides the first description of the primordial follicular oocyte proteome and further illustrates the dynamic shifts in protein abundance associated with chemotherapeutic agents, laying the foundation for further development of effective and targeted drugs to protect and prolong female fertility.

Proteomic analysis for Cisplatin -induced mice spermatogenesis defects

Project description:Cisplatin is a crucial chemotherapeutic agent for treating various cancers, but excessive use can cause irreversible damage to the reproductive system. However, the protein expression profile of cisplatin-induced testicular injury remains poorly understood. In this study, we observed that cisplatin treatment resulted in smaller testes, reduced sperm count, and a significant decrease in the number of spermatocytes and spermatids in mice. Further label-free quantitative proteomic analysis revealed that cisplatin reduced the expression of CDK1 in the testes, thereby affecting the number of spermatocytes and spermatids. These findings provide new insights into fertility preservation for cancer patients undergoing chemotherapy.

MEK Inhibition Overcomes Cisplatin Resistance Conferred by SOS/MAPK Pathway Activation in Squamous Cell Carcinoma (SCC)

Project description:Genomic analyses of SCC have yet to yield significant strategies against pathway activation to improve treatment. Platinum-based chemotherapy remains the mainstay of treatment for SCC of different histotypes either as a single agent or alongside other chemotherapeutic drugs or radiotherapy; however, resistance inevitably emerges, which limits the duration of treatment response. To elucidate mechanisms that mediate resistance to cisplatin, we compared drug-induced perturbations to gene expression between cisplatin-sensitive and -resistant SCC cells. Cisplatin-sensitive and -resistant SCC cells were identified through MTS assay. Gene expression profiling (Affymetrix GeneChip 1.0ST) was performed to identify putative genes and cellular pathways that may be associated with cellular response to cisplatin.

Istradefylline (KW6002) protects from cisplatin-induced nephrotoxicity and peripheral neuropathy while preserving cisplatin anti-tumor effects [Kidney_Tumor]

Project description:Cisplatin is a potent chemotherapeutic drug, widely used in the treatment of various solid cancers. However, its clinical effectiveness is strongly limited by frequent severe adverse effects, such as nephrotoxicity and chemotherapy-induced peripheral neuropathy. Therefore, there is an urgent medical need to identify novel strategies that would limit cisplatin-induced toxicity. In the present study, we provide evidence that the FDA-approved adenosine A2A receptor antagonist istradefylline (KW6002) significantly protects from cisplatin-induced nephrotoxicity and neuropathic pain in experimental models of acute and sub-chronic cisplatin intoxication. Importantly, we also demonstrate that the anti-tumoral properties of cisplatin were not altered by istradefylline in tumor-bearing mice and even potentiated at the molecular level. Altogether, the present results support the use of istradefylline as a new valuable preventive approach for the clinical management of patients undergoing cisplatin treatment.
