Project description:Blood vessels show various COVID-19-related conditions including thrombosis and cytokine propagation. Existing in vitro blood vessel models cannot represent the consequent changes in the vascular structure or determine the initial infection site, making it difficult to evaluate how epithelial and endothelial tissues are damaged. Here, we developed a microphysiological system (MPS) that co-culture the bronchial organoids and the vascular bed to analyze infection site and interactions. In this system, virus-infected organoids caused damage in vascular structure. However, vasculature was not damaged or infected when the virus was directly introduced to vascular bed. The knockout of interferon-related genes and inhibition of the JAK/STAT pathway reduced the vascular damage, indicating the protective effect of interferon response suppression. The results demonstrate selective infection of bronchial epithelial cells and vascular damage by cytokines and also indicate the applicability of MPS to investigate how the infection influences vascular structure and functions.

Project description:The study of cardiac physiology and disease is hindered by physiological differences between humans and small-animal models. Here, we report the generation of multi-chambered vascularized human cardiac organoids under anisotropic stress, and their applicability to study electro-metabolic coupling in cardiac tissue. The organoids are derived from human induced pluripotent stem cells, and integrate sensors for the simultaneous measurement of oxygen uptake, extracellular field potentials and cardiac contraction at resolutions higher than 10 Hz. The microphysiological system allowed us to find that 1-Hz cardiac respiratory cycles are coupled with electrical activity rather than with mechanical activity, that calcium oscillations drive a mitochondrial respiration cycle, that the pharmaceutical or genetic inhibition of electro-mitochondrial coupling results in arrhythmogenic behaviour, and that the induction of arrythmia by the chemotherapeutic mitoxantrone can be partially reversed by the co-administration of metformin. Microphysiological cardiac systems may further facilitate the study of the mitochondrial dynamics of cardiac rhythms and advance the understanding of cardiac physiology.

Project description:We combined human induced pluripotent stem cells (hiPSCs) and 3D printing to mass-produced liver Disse space-like organoids in microspheres hydrogels (DOs). After 5 days of co-culture, the cells within the microspheres were aggregated and developed into liver organoids, composed of iPSC-derived hepatocytes, endothelial cells, and hepatic stellate cells (LX2). These organoids mirrored the hepatotoxicity of polystyrene microplastics (PS-MPs), TBBPA, and complexes MPTA with healthy and patient-derived organoids.

Project description:Ochratoxin A (OTA) is one of the most abundant mycotoxin contaminants in food stuffs and possesses carcinogenic, nephrotoxic, teratogenic and immunotoxic properties. Especially, severe nephrotoxicity is of great concern, as characterized by degeneration of epithelial cells of the proximal tubules and interstitial fibrosis. However, its mechanism of toxicity, hazard identification as well as genetic risk factors contributing to OTA toxicity in humans has been elusive due to the lack of adequate models that fully recapitulate kidney function in vitro. The present study attempts to evaluate dose-response relationships, identify the contribution of active transport proteins that govern renal disposition of OTA, and determine the role of metabolism in bioactivation and detoxification of OTA using a 3D human kidney proximal tubule microphysiological system (kidney MPS). We demonstrated that IC50 values of OTA in kidney MPS culture (0.375 – 1.21 µM) were in good agreement with clinical toxic concentrations of OTA in urine. Surprisingly, no enhancement of kidney injury biomarkers was evident in the effluents of kidney MPS after OTA exposure despite significant toxicity observed by LIVE/DEAD staining, rather these biomarkers were decreased in OTA concentration-dependent manner. Furthermore, the effect of 1-aminobenzotriazole (ABT) and 6-(NBD-4-ylthio-) hexanol (NBDHEX), pan-inhibitor of P450 and GST enzymes, respectively, on the OTA-induced toxicity in kidney MPS was examined, which resulted in significant enhancement of OTA-induced toxicity by NBDHEX (3 µM) treatment whereas ABT (1 mM) treatment decreased OTA-induced toxicity, suggesting the roles of GSTs and P450 enzymes in the detoxification and bioactivation of OTA, respectively. Additionally, OTA transport studies using kidney MPS in the presence and absence of inhibitor of organic anion transporter(s), probenecid (1 mM), revealed the role of organic anionic membrane transporter(s) in the kidney specific disposition of OTA. Our findings provide a better understanding of the mechanism of OTA-induced kidney injury which may support changes in risk assessment, regulatory agency policies on allowable exposure levels and determination of genetic factors in high-risk populations against OTA nephrotoxicity.

Project description:Current research on metabolic disorders and diabetes relies on animal models because multi-organ diseases cannot be well studied with the standard in vitro assays. Here, we connect models of key metabolic organs, pancreas and liver, on a microfluidic chip to enable diabetes research in a human-based in vitro system. Aided by mechanistic mathematical modelling, we show that hyperglycemia and high cortisone induce glucose dysregulation in the pancreas-liver microphysiological system (MPS) mimicking a diabetic phenotype seen in patients with glucocorticoid-induced diabetes. In this diseased condition, pancreas-liver MPS displays beta-cell dysfunction, steatosis, elevated ketone-body secretion, increased glycogen storage, and upregulated gluconeogenic gene expression. In turn, a physiological culture condition maintains the glucose tolerance and beta cell function. This method was evaluated for reproducibility in two laboratories and was effective in multiple pancreatic islet donors. The model also provides a platform to identify new therapeutic proteins as demonstrated with a combined transcriptome and proteome analysis.

Project description:Drug-induced liver injury (DILI) remains a major challenge in drug development and inter-species differences in potential liver toxicity are one area where comparative analyses may inform pre-clinical safety study design. In vitro testing for species-specific liver effects, especially in complex models such as microphysiological systems (MPS) may help predict toxicity before advancing from animal to human studies or de-risk spurious findings in pre-clinical species. This study assessed the utility of the perfusion-based PhysioMimix® LC12 MPS for evaluating species-specific DILI using primary hepatocytes from human, monkey, rat, and dog. First, functional, phenotypic, and transcriptional profiles were established over 2 weeks in cell culture. Then, cells were exposed to species-specific DILI compounds – chlorpromazine (CPZ), bosentan (BOS), and fialuridine (FIAU) – in both PhysioMimix® LC12 and traditional 2D cultures. Hepatocytes in PhysioMimix® LC12 showed more stable albumin and urea production for up to 14 days as compared to 2D cultures. Concentration-response studies with CPZ, BOS, or FIAU were performed in 2D; then, prolonged exposures (10 days) to sub-100× Cmax concentrations were tested in PhysioMimix® LC12. Species-specific differences in cellular and molecular effects of the drugs were observed in both models; data from PhysioMimix® LC12 were more reflective of the expected effects in whole animals or humans. Still, variability and low throughput are limitations of the MPS for prospective studies of species-specific responses. Overall, this study confirms the utility of liver safety studies using MPS, but also provides suggestions for experimental designs to overcome the limitations of more complex test systems.

Project description:In this study, we demonstrate the use of human cardiac organoids (comprised of cardiomyocyte, fibroblast, and endothelial origin) to model IR injury through a model of hypoxia and reoxygenation and therapeutic nanovesicle remodelling. Engineered nanovesicles (NVs), generated directly from human stem cells (SC), have been shown to influence cardiac tissue repair, and provide a platform for the reproducible, rapid, and scalable cell free-mediated therapy. Functionally, we demonstrate that administration of NVs (from different human induced pluripotent stem cell (iPSC) origin) during reoxygenation significantly increase cardiomyocyte survival and preserve contractility function (contractile duration, relaxation time, relaxation:contraction velocity). A mass-spectrometry-based proteomics approach was applied to decipher protein dynamics and molecular mechanisms of IR injury in human cardiac organoids following NV treatments
