Analysis of gene expression in irradiated intratumoral mouse T cells

ABSTRACT: We aimed to identify promoters activated by ionizing radiation (IR) specifically in T cells by analyzing gene expression in intratumoral mouse T cells after in vivo IR treatment . Whole-body irradiation (WBI) was used instead of local IR to avoid contamination by non-irradiated T cells newly infiltrating from the periphery. MC38 tumor-bearing B6 mice received 1 or 5 Gy WBI, and differential gene expression was analyzed in FACS-sorted T cells from tumors at 1 h, 6 h, or 24 h post-irradiation, vs. T cells sorted from non-irradiated tumors.
