Effect of Sox2-activation on transcriptomic profiles of murine lung tumors

ABSTRACT: To investigate the effect of Sox2-activation during lung tumorigenesis, we treated CRISPRa competent PPKS (KRAS(LSL-G12D//wt); TP53(flox/flox); Rosa26(LSL-SAM/wt)) mice with CMV-Cre/U6-sgRNA(MS2) encoding lentivirus carrying Sox2-targeted or non-targeted sgRNA. Tumors were harvested at survival endpoint and transcriptomic reprogramming was assessed with RNA-seq.

In vivo CRISPR activation screening of tumor formation in murine pancreas and lung.

Project description:We performed in vivo CRISPR activation screening of tumor formation in the pancreas and lung by treating CRISPRa competent PPKS (KRAS(LSL-G12D//wt); TP53(flox/flox); Rosa26(LSL-SAM/wt)) mice with CMV-Cre/U6-sgRNA(MS2) lentivirus containing the Onco1 sgRNA guide library. Tissues were harvested at different timepoints (6 days, 4 weeks, 8 weeks and survival endpoint). HEK293T cell cultures were transduced with the same libraries to generate normalization controls. Library plasmid DNA (pDNA) was sequenced as controls.

Expression profiling of Notch constitutive activation in the liver of mice

Project description:Notch intracellular domain (NICD) is the active form of the Notch receptor. In this mouse model, NICD is inserted in the Rosa26 locus downstream of a loxP-STOP-LoxP (lsl) sequence and therefore NICD expression is dependant on Cre recombinase expression (Mono transgenic control Rosa26-lsl-NICD) . These mice are crossed with the AFP-Cre strain that expresses Cre in hepatoblasts due to its regulation by the AFP promoter and albumin enhancer (Double transgenic mutant AFP-Cre/Rosa26-lsl-NICD). Newborn mice at day 0 and day 2 are sacrificed and liver RNA samples from control monotransgenic Rosa26-lsl-NICD and from bitransgenic AFP-Cre/Rosa26-lsl-NICD (AFP-NICD) are obtained. Whole genome expression profiling of these samples is submitted.

Impact of IRP deficiency on the proteome of Ly6G+ cells from the bone marrow in mice

Project description:The overall objective of the project is to evaluate the effects of loss of function of iron regulatory proteins (IRP1, encoded by Aco1)-1 and -2 (IRP2, encoded by Ireb2) during adult life. Mice carrying floxed Aco1 and Ireb2 alleles were crossed to a knockin strain expressing a tamoxifen-inducible CRE recombinase under the control of the Rosa26 promoter. The resulting Aco1flox/flox,Ireb2flox/flox,Rosa26+/CreERT2 mice are designated P1/2-KO; littermates lacking the CreER sequence (Aco1flox/flox,Ireb2flox/flox,Rosa26+/+) serve as reference and are designated P1/2-CTR. Adult mice were injected (i.p.) with tamoxifen on day 1 and day 3 to ablate the IRPs in the whole body, and were sacrificed on day 10 for analysis. Bone marrow cells were collected and LY6G+ cells were magnetically sorted for proteome analysis by LC-MS/MS using an Ultimate 3000 UPLC system connected to an Orbitrap Exploris 480 mass spectrometer.
