DNA Methylation Profiles Predicting Response to Anti-PD-1-based Treatment in Patients with Advanced Gastric Cancer

ABSTRACT: Immunotherapy has shown promise in treating gastric cancer, but predicting patient response remains challenging. In this study, DNA methylation profiles from 30 samples were analyzed using the Infinium MethylationEPIC BeadChip (850K array). A total of 523 differentially methylation probes (DMPs) were identified, from which 20 CpG probes were selected as the most predictive of treatment response. Based on these 20 DMPs, we developed the iMETH model, which accurately predicts patient responses to first-line anti-PD-1-based therapy.

Project description:Immunotherapy has shown promise in treating gastric cancer, but predicting patient response remains challenging. In this study, DNA methylation levels at 20 specific CpG probes were measured in 69 tumor samples using targeted bisulfite sequencing (TBS). These CpG probes, previously identified as highly predictive of treatment response, were used to construct and validate a methylation-based prediction model. The resulting iMETH model accurately stratified patients according to their likelihood of responding to first-line anti-PD-1-based therapy.

In this study, we have investigated the effect of pregnancy on circulating CD4+ and CD8+ T cells from patients with multiple sclerosis and healthy controls to gain a deeper understanding why patients with multiple sclerosis improves during pregnancy. We assessed epigenome-wide DNA methylation in CD4+ and CD8+ T cells obtained during (1st, 2nd and 3rd trimester) and after pregnancy (6 weeks post-partum), using the Infinium MethylationEPIC 850K array.
