Single-nucleus RNA-seq of peripheral blood mononuclear cells from multiple sclerosis patients and healthy controls

ABSTRACT: Multiple sclerosis (MS) is a chronic immune-mediated disease of the central nervous system marked by neuroinflammation and demyelination. Emerging evidence suggests that circulating immune cells contribute to disease activity, heterogeneity of clinical course, and treatment response. Here we provide single-nucleus transcriptomes of peripheral blood mononuclear cells (PBMCs) from individuals with MS and healthy controls. The dataset enables cell-type–resolved interrogation of peripheral immune states associated with MS, including comparisons of cell-state frequencies and gene-expression programs between groups, exploration of inflammatory pathways and ligand–receptor interactions, and discovery of candidate biomarkers. This resource supports integrative analyses with external MS cohorts and facilitates hypothesis generation on mechanisms of immune dysregulation in MS.

Interleukin-3 coordinates glial-peripheral immune crosstalk to incite multiple sclerosis

Project description:Multiple sclerosis (MS) is a neuroinflammatory disease of the central nervous system (CNS). While CNS-resident glial cells and infiltrating immune cells contribute to MS pathogenesis, the networks that govern peripheral-central immune crosstalk and coordinate functionality among these ontologically distinct populations remain unclear. Here we show, in mice and humans, that astrocyte- and CD44hiCD4+ T cell-sourced interleukin-3 (IL-3) programs microglia and infiltrating myeloid cells to exacerbate neuroinflammation by inciting a cellular recruitment program that drives the accrual of immune cells in the CNS thereby worsening MS and its preclinical model experimental autoimmune encephalomyelitis (EAE). We find that CNS-resident astrocytes and peripherally-derived CD44hiCD4+ T cells generate IL-3 while resident microglia and infiltrating monocytes, macrophages, and dendritic cells respond to the cytokine by expressing IL-3Rɑ, IL-3's specific receptor. Astrocytic and T cell IL-3 elicit an immune migratory, recruitment, and chemotactic program by IL-3Rɑ+ myeloid cells that enhances immune cell infiltration into the CNS leading to exacerbated neuroinflammation, demyelination, and clinical disease. Multiregional single-nuclei RNA sequencing (snRNAseq) of human CNS tissue from unaffected controls and MS patients reveals the appearance of a subset of IL3RA-expressing myeloid cells in MS plaques with unique recruitment, migratory, and chemotactic programing and function. In humans with MS, IL3RA expression by plaque myeloid cells and IL-3 levels in the cerebrospinal fluid (CSF) predict myeloid and T cell abundance and recruitment into the CNS. Together, our findings establish IL-3:IL-3RA signaling as a bidirectional glial-peripheral immune crosstalk network that promotes neuroinflammation, prompts immune cell recruitment to the CNS, and worsens MS.

Suppression of 6-pyruvoyl-tetrahydropterin synthase promotes remyelination in multiple sclerosis

Project description:Background: Multiple sclerosis (MS) is a CNS autoimmune disease that is characterized by demyelination, neuroinflammation, and neuronal loss. Although current chemotherapies and immunotherapies for MS efficiently mitigate disease activity and relief clinical symptoms, neuroprotection targeting axon/neurons remains therapeutically challenging. Accumulating evidence has suggested that poor oligodendrocyte precursor cell (OPC) proliferation and subsequent migration/differentiation significantly impedes remyelination during MS. However, the molecular mechanisms underlying OPCs dysfunction during MS remain elusive. Methods: Re-analysis of single cell sequencing data from human and mouse oligodendrocyte lineage cells (OLCs), along with staining results from brain sections of MS patients, was conducted to investigate the role of 6-pyruvoyl-tetrahydropterin synthase (PTS) in determining oligodendrocyte fate. To assess PTS function in MS, conditional knockout mice targeting OLCs were employed. Transcriptomic analyses further illuminated the molecular mechanisms through which PTS influences the disease process. Results: PTS in OPCs is a suppressor of OPC proliferation that contributes to demyelination in MS. We found that PTS expression in OPCs was aberrantly increased in patients with MS. Mice lacking Pts specifically in OPCs displayed ameliorated experimental autoimmune encephalitis (EAE) severity, a mouse model of MS, which were accompanied by attenuated demyelination. Moreover, selective suppression of Pts in OPCs facilitated OPC proliferation and remyelination during EAE. Mechanistically, the knockdown of Pts activated the cholesterol biosynthesis pathway leading to enhanced OPC proliferation. Conclusions: Our findings suggest that PTS is a negative regulator of OPC proliferation, and its disinhibition could offer a potential therapeutic target for MS.

Oligodendroglial precursors orchestrate immune cellular network instigating multiple sclerosis

Project description:The immunomodulatory cellular network that triggers early inflammation and demyelination, the key steps in multiple sclerosis (MS) pathogenesis remains poorly characterized. Here, we demonstrate that overactivation of the Wnt pathway promotes pathological transformation of oligodendrocyte precursor cells (OPCs) to replicate pathological OPCs in human MS. In mouse experimental autoimmune encephalomyelitis (EAE), pathological OPCs attract CD4+ T-helper 1 (Th1) cells into the spinal cord and the brain through CC-chemokine ligand 4 (CCL4). Th1 cells cooperate with OPCs inducing subpopulation of cytotoxic macrophages that execute early demyelination. Simultaneously, Th1 cells and cytotoxic macrophages upregulate Wnt signaling and CCL4 expression in OPCs, thus exerting positive feedback onto the OPC-immune cascade and establishing a vicious cycle propagating EAE pathogenesis. Breaking this cascade by targeting CCL4 reduces immune cell infiltration, alleviates demyelination, and attenuates EAE severity. Our findings demonstrate a closely coordinated network of OPCs and immune cells therefore providing an alternative insight into MS pathophysiology

Oxidative damage and respiratory burst in Multiple Sclerosis

Project description:In this project we focused on white matter injury in relapsing remitting MS. We concentrated on material obtained from patients who suffered from fulminant active MS to identify possible sources for ROS production in relation to demyelination and neurodegeneration. To identify possible sources for ROS production in relation to demyelination and neurodegeneration in MS, we used genome wide microarray analysis of gene expression in carefully dissected lesion areas of patients with fulminant acute MS. Preceding the gene expression profiling all cases were characterized histologically and areas of interest were identified. We included 4 control cases, 2 active MS cases were we separated 3 distinct lesions areas (NAWM, initial lesion and early demyelinated lesion) and 1 active MS case were we separated 2 distinct lesion areas (NAWM and early demyelinated lesion). The RNA was isolated from those areas, amplified and hybridized to Agilent G4112A whole genome microarrays.

Disease modifying therapy impacts on miRNA expression profiling in peripheral blood from Relapsing-Remitting Multiple Sclerosis patients

Project description:Multiple sclerosis (MS) is a chronic and progressive neurological disease. MS is characterized by early-stage neuroinflammation, neurodegeneration, and demyelination, with a spectrum of heterogeneous clinical manifestations in terms of disease course and response to therapy. The currently unsatisfied availability of diagnostic and prognostic biomarkers concerning disease progression and treatment response represents an important requirement for therapy individualization and drug efficacy. Specific Disease Therapies (DMTs) are available to prevent MS-related brain damage; however, the specific drug choice is still under debate and needs further characterization. Since differentially expressed miRNAs have been proposed as diagnostic tools in neurodegenerative/neuro-inflammatory diseases, such as multiple sclerosis, amyotrophic lateral sclerosis, and Alzheimer's disease, we characterized the miRNA expression profiling in peripheral blood mononuclear cells (PBMC) of subjects with relapsing-remitting MS treated with high efficacy DMTs: Cladribine (CLA, n=11 patients) or Ocrelizumab (OCRE, n=14 patients). The treated patients were compared to control (CTR) untreated subjects (n=15). Blood samples were collected from patients before treatment (time t0) and 6 months post-treatment (time t1), but just once from control subjects. CLA drug tablets were administered to the selected relapsing-remitting MS patients with a dosage of 1.75 mg/kg given for 5 days. OCRE drug was administered to the other relapsing-remitting MS patients by infusion with a dosage of 300 mg, twice in two weeks.

Transcription signature of Multiple Sclerosis in peripheral blood mononuclear cells.

Project description:Background: pregnancy is associated with reduced activity of multiple sclerosis (MS). However, the biological mechanisms underlying this pregnancy-related decrease in disease activity are poorly understood. This data series contains the subset of data used to generate a MS signature comparing female healthy specimens with respect to MS patients Subjects were followed in the outpatients clinic and blood was collected before pregnancy and at the following time points during pregnancy: first trimester (gestational age at sampling 12 weeks), second trimester (24 weeks), and third trimester (36 weeks). Before-pregnancy samples were obtained in a treatment-free period and after anticonceptional drug withdrawal. Peripheral blood mononuclear cells (PBMCs) obtained from 15 women (8 MS patients and 7 healthy controls) were analyzed by oligonucleotide microarray technology.

Consequences of perinatal bisphenol A exposure in a mouse model of multiple sclerosis

Project description:Multiple sclerosis (MS) is a complex disease influenced by genetic and environmental contributing factors. Endocrine disrupting compounds (EDCs) such as bisphenol A (BPA) affect gene expression and hormone-regulated systems throughout the body. We investigated the effects of BPA on Theiler’s-virus induced demyelination (TVID), a mouse model of MS. Perinatal BPA exposure, combined with viral infection, resulted in a decreased level of viral antibodies, accelerated the onset of TVID symptoms, increased inflammation in both the spinal cord and digestive tract, and amplified immune-related gene expression changes induced by viral infection. These results demonstrate the effect of BPA on the trajectory of TVID, and illustrate how multiple factors collectively influence autoimmune disease. For microarray: 7 samples
