DNA Methylation Profiles Predicting Response to Anti-PD-1-based Treatment in Patients with Advanced Gastric Cancer [Targeted Bisulfite-Seq]

ABSTRACT: Immunotherapy has shown promise in treating gastric cancer, but predicting patient response remains challenging. In this study, DNA methylation levels at 20 specific CpG probes were measured in 69 tumor samples using targeted bisulfite sequencing (TBS). These CpG probes, previously identified as highly predictive of treatment response, were used to construct and validate a methylation-based prediction model. The resulting iMETH model accurately stratified patients according to their likelihood of responding to first-line anti-PD-1-based therapy.
