Cellular stress-induced eccrine gland dysfunction as a potential mechanism in acquired idiopathic generalized anhidrosis

ABSTRACT: Acquired idiopathic generalized anhidrosis (AIGA) is a rare disorder characterized by systemic anhidrosis or hypohidrosis of unknown etiology. Corticosteroid pulse therapy is widely used for its treatment. We analyzed skin lesions from AIGA patients before and after corticosteroid pulse therapy through single-cell RNA sequencing analyses.

Project description:Eccrine sweat glands contribute to the regulation of human body temperature. Acquired idiopathic generalized anhidrosis (AIGA) is extensive anhidrosis of unknown etiology, that causes the fatal heatstroke and skin dryness. Eccrine sweat glands in hidrotic and anhidrotic skin areas were excised from paraffin-embedded skin specimens from AIGA patients, and the gene expression was profiled by RNA-sequencing. We analyzed the RNA samples to clarify the difference in gene expression between hidrotic and anhidrotic skin areas from AIGA patients.

Gene expression changes associated with resistance to intravenous corticosteroid therapy in children with severe ulcerative colitis

Project description:Although corticosteroids remain a mainstay of therapy for UC, a meta-regression of cohort studies in acute severe ulcerative colitis (UC) showed that 29% of patients fail corticosteroid therapy and require escalation of medical management or colectomy. We aimed to determine whether genes expressed in whole blood early following initiation of intravenous corticosteroid treatment can be associated with response. 20 corticosteroid responsive (PUCAI < 35 by day 5) and 20 corticosteroid resistant patients (PUCAI ≥ 35 by day 5 and need for 2nd line medical therapy or colectomy by hospital discharge) were selected from a prospectively accrued cohort of patients hospitalized for intravenous corticosteroid treatment of severe UC. Total RNA was extracted from blood samples collected on day 3 of intravenous corticosteroid therapy. The eluted transcriptomes were quantified on Affymetrix Human Gene 1.0 ST arrays. The data was analysed by the local-pooled error method for discovery of differential gene expression and false discovery rate correction was applied to adjust for multiple comparisons. P-values less than 0.05 after correction were considered significant.

VitGene Generalized Vitiligo Genetics Study

Project description:Generalized vitiligo is an autoimmune disease in which melanocyte loss results in patchy depigmentation of skin and hair. For this GWAS of generalized vitiligo, 579,146 SNPs were genotyped in 1514 generalized vitiligo cases of European-derived white (CEU) ancestry versus data from 2813 "public" CEU controls. 49 strongly associated SNPs from the first phase of the GWAS were then tested in two replication studies, one comparing 677 independent cases and 1106 CEU controls, and the other a family-based analysis of 204 CEU simplex generalized vitiligo trios and 310 CEU multiplex families. Overall, significant association of generalized vitiligo was detected for SNPs in several genes previously associated with other autoimmune diseases, including MHC class I (P = 9.05 x 10-23) and class II (P = 4.50 x 10-34) loci, PTPN22 (P = 1.31 x 10-7), LPP (P = 1.01 x 10-11), IL2RA (P = 2.78 x 10-9), UBASH3A (P = 1.26 x 10-9), and C1QTNF6 (P = 2.21 x 10-16). Association was also detected for SNPs in two novel immune-related loci, RERE (P = 7.07 x 10-15), GZMB (P = 3.44 x 10-8), and in a locus that may mediate melanocyte target cell specificity, TYR (P = 1.60 x 10-18) encoding tyrosinase. These findings thus demonstrate association between generalized vitiligo and multiple genes, some associated with other autoimmune diseases and others that may mediate target-cell specificity, and suggest a possible inverse relationship between susceptibility to vitiligo versus melanoma.
