ScRNA-seq analysis of liver nonparenchymal cells from the mouse model of CCl4-induced liver fibrosis

ABSTRACT: We performed scRNA-seq analysis on livers from the mouse model of CCl4-induced liver fibrosis. The scRNA-seq analysis showed that CTGF expression was increased only in zone 2 LSECs, and that YAP was not activated in any cluster of LSECs.

Project description:We performed scRNA-seq analysis on murin livers after partial inferior vena cava ligation (pIVCL), a mouse model of congestive hepatopathy. We identified connective tissue growth factor (CTGF) as one of the most upregulated genes in pericentral (zones 2 and 3) LSECs. The scRNA-seq analysis also showed that the integrin signaling pathway was activated and the expression of YAP/TAZ target genes was upregulated in pericentral LSECs.

Project description:We performed scRNA-seq analysis on the livers from the endothelial cell-specific CTGF knockout (KO) mouse or control wild-type (WT) littermate after partial inferior vena cava ligation (pIVCL), a mouse model of congestive hepatopathy. The scRNA-seq analysis showed that CTGF knockout in LSECs suppressed the expression of YAP/TAZ target genes and basement membrane-related genes in LSECs caused by hepatic congestion.

MicroRNA expression profiling of CCl4-induced liver fibrosis in Mus musculus

Project description:To investigate the differences in microRNA expression profiles between fibrotic and normal livers, we performed microRNA microarrays for total RNA extracts isolated from mouse livers treated with carbontetrachloride (CCl4) or corn-oil for 10 weeks (n=3/group). MicroRNAs were considered to have significant differences in expression level when the expression difference showed more than two-fold change between the experimental and control groups at p<0.05. We found that 12 miRNAs were differentially expressed in CCl4-induced fibrotic liver. To induce chronic liver fibrosis, seven-week-old mice received 0.6 ml/kg body weight of carbon-tetrachloride (CCl4) dissolved in corn-oil by intraperitoneal (i.p.) injection, twice a week for 10 weeks (n=3). As a control, same number of mice was injected with equal volume of corn-oil for 10 weeks.

Next Generation Sequencing (NGS) Facilitates Quantitative Analysis of Differentially Expressed Genes in Liver Regeneration After Combined Treatment With Ethanol And Carbon Tetrachloride

Project description:Purpose: The goals of this study are to use NGS-derived liver transcriptome profiling (RNA-seq) and identify differentially expressed genes in regenerating livers that were treated with ethanol and carbon tetrachloride. Methods: Liver mRNA profiles were generated from ethanol/CCl4-treated floxed homozytoes and Ctgf conditional knockout mice by deep sequencing, in duplicate, using Illumina GAIIx. qRT–PCR validation was performed using SYBR Green assay. Results: Ctgf deficiency was associated with 220 downregulated genes and 29 upregulated genes whereas Yap1 null livers had 351 downregulated genes and 287 upregulated genes after ethanol/CCl4 induced liver damage Conclusions: This study provides detailed analysis of liver transcriptomes in absence of Ctgf or Yap1 genes during ethanol/CCl4-induced injury, with biologic replicates, generated by RNA-seq technology.
