Transcription profiling of Drosophila melanogaster third instar larvae organ cultures treated with 20-hydroxyecdysone, cycloheximide vs controlds to identify 20-hydroxyecdysone regulated genes at the onset of metamorphosis

Project description:To identify 20E-regulated genes, wandering third instar larvae were dissected and their organs were cultured in the presence of either no hormone, 20E alone, cycloheximide alone, or 20E plus cycloheximide for six hours. Experiment Overall Design: Eight partial blue gut third instar larvae were dissected in each well of a 9 well glass dish (Corning) and cultured in ~100 Âµl oxygenated Schneiders Drosophila Medium (Gibco) at 25Â°C. Cultures were incubated in an a styrofoam box under a constant flow of oxygen. Following an initial incubation of 1 hour, the medium was removed and replaced with either fresh Schneiders Drosophila Medium (no hormone), medium plus 8.5x10-5 M cycloheximide (Sigma), medium plus 5x10-6 M 20-hydroxyecdysone (Sigma), or medium plus cycloheximide and 20E, each for 6 hrs at 25Â°C. Organs were collected and RNA extracted from these samples was analyzed on Affymetrix Drosophila Genome Arrays.

Expression profile of third instar larval fat body and midgut tissues

Project description:We use mRNA-seq to transcriptionally profile larval fat body and midgut tissues from Drosophila third instar larvae. These data provide insights into tissue physiology and can be used to identify tissue specific transcripts. Fat bodies from wandering third instar larvae were dissected from ~50 male larvae and gonads were removed to eliminate contaminating transctips from the gonads. Larval midguts were dissected from ~50 wandering third instar larvae. Larval tissues were removed to Graces unsupplemented medium on ice prior to RNA extraction with TRIzol reagent. mRNA-seq samples were prepared from 5ug of total RNA and subject to Illumina based sequencing.

Genome-wide examination of the transcriptional response to ecdysteroids 20-hydroxyecdysone and ponasterone A in Drosophila melanogaster

Project description:The 20-hydroxyecdysone (20E) hierarchy of gene activation serves as an attractive model system for studying the mode of steroid hormone regulated gene expression and development. Primary genes are activated by the direct action of the hormone and often code for proteins with regulatory function, some of which are responsible for the perpetuation of the response through the activation of secondary genes. Here we identify 35 primary 20E-response genes that are induced by 20E in the absence of protein synthesis in the embryonic cell line Kc167, and of those with known functions, most encode proteins with catalytic activity. Comparison of the genome-wide transcriptional response to 20E and to its plant derived structural analog ponasterone A (PoA) revealed a large difference in the transcriptional targets of these molecules. Despite the structural similarity of these two compounds, many more genes related to various aspects of development appear to be significantly induced by 20E than by PoA. Furthermore, we compared the 20E response in Kc cells to that of a natural 20E target tissue where the function of 20E has been well described, the salivary glands of wandering 3rd instar larvae, and found little overlap in 20E-responsive genes. Analysis of 20E-induced transcription of EcR, a known 20E-inducible gene and regulator of early and late 20E-responsive genes, reveals differences in the fold induction of EcR isoforms EcR-RA, ER-RC, and EcR-RD/E between Kc cells and salivary glands suggesting a possible cause for the observed differences in 20E-regulated gene transcription.

20-hydroxyecdysone (20E) induced transcriptional changes in the atrium and spermatheca of female Anopheles gambiae mosquitoes 24 hours after 20E injection compared to ethanol treated controls.

Project description:Male Anophele gambiae mosquitoes transfer the steroid hormone 20-hydroxyecdysone (20E) to the female lower reproductive tract during mating. We have shown that this sexually transferred hormone is a key regulator of monandry and oviposition in An. gambiae females. In order to identify possible molecular pathways underlying the effects induced by sexually transferred 20E we investigated the transcriptional response in the atrium and spermatheca at 24 hours after 20E injection in the thorax of virgin females, which delivers to these reproductive tissues 20E levels comparable to those detected after mating. By comparing these results to those of microarrays assessing the post-mating transcriptional response in the female lower reproductive tract (LRT) at 24 hours post copulation, we were able to identify mating responsive genes likely regulated by male transferred 20E. For each experiment atrium and spermatheca were dissected from females 24 hours after 20E or ethanol (10%) injection and transcriptional profiles compared across 4 biological replicates using Agilent two-color microarrays

Transcriptomic analysis of Drosophila imaginal wing discs cultured in insulin or 20-hydroxyecdysone

Project description:We identified a role for low concentrations of 20-hydroxyecdysone (20E) in promoting growth of the Drosophila larval wing imaginal disc using genetics and in vitro explant culture. We found that 20E alone could promote proliferation in the absence of added insulin, a well known regulator of growth. To investigate the mechanism by which these hormones promote growth, we sequenced the transcriptomes of mid-third instar wing discs cultured in no hormone, 20E alone or insulin alone at two timepoints (4hr and 9hr). We also sequenced the transcriptomes of freshly dissected wing discs (not cultured). We found that within 4hr of culture without hormone, the expression of 1604 genes goes down and 1409 genes goes up, relative to uncultured discs. Changes in the expression of 1911 genes could be prevented, at least transiently, by the addition of either 20E or insulin. The transcriptional response to insulin was remarkably transient, decreasing by rougly 50% from 4hr to 9hr of culture. The response to 20E was smaller than that induced by insulin, but longer lasting. Insulin-induced genes are most significantly enriched in GO-terms describing DNA replication, ribosome biogenesis, translation, purine biosynthesis, and mitochondrial biogenesis. Insulin-repressed genes are enriched in genes involved in autophagy and cell death. 20E-regulated genes were also enriched for genes involved in DNA replication. Unlike insulin, however, 20E was found to promote the expression of genes in GO categories describing a variety of developmental processes. These data represent the first description of the direct transcriptional response of the wing disc to either insulin or low doses of 20E.

Expression data from 0.15M and 0.7M-fed wild-type and ChREBP mutant, third instar Drosophila larval fat bodies (FBs)

Project description:Chronic high sugar feeding induces obesity, hyperglycemia, and insulin resistance in flies and mammals. These phenotypes are controlled by the fat body, a liver- and adipose- like tissue in Drosophila flies. To gain insight into the mechanisms underlying the connection between diet and insulin sensitivity, we used Illumina RNA-seq to profile gene expression in fat bodies isolated from chronically high sugar fed, wandering (post-prandial) third instar wild type larvae w(L3). These data were compared to control-fed wild-type wL3 fat bodies as well as those expressing transgenic interfering RNA (i) targeting CG18362 (Mio/dChREBP) in the fat body on both diets. Female VDRC w1118, cgGAL4, UAS-Dcr2 or UAS-ChREBPi(52606), cgGAL4, UAS-Dcr2 wandering third instar larvae were fed control (0.15M) or high (0.7M) sucrose and fat bodies isolated for RNA extraction.
