Project description:Tumor-draining lymph nodes (TdLNs) were collected from wild-type (WT) and tenascin-C knockout (TNC-KO) C57BL/6 mice (N = 3 per group), each bearing 4NQO-induced tongue tumors. Both male and female mice were included. Beginning at 12 weeks of age, mice were exposed to 4NQO via drinking water for 16 weeks. Tumors were then subjected to a single 2 Gy irradiation. TdLNs were harvested six weeks after irradiation, and total RNA was isolated for bulk RNA-seq analysis.

Project description:Regional lymph node metastasis critically influences distant metastatic progression, anti-tumour immunity, and patient prognosis. This study systematically characterised immune cell dynamics across paired primary tumours, sentinel tumour-draining lymph nodes (S-TDLNs), and secondary axillary lymph nodes (ALNs) from treatment-naïve triple-negative breast cancer (TNBC) patients using integrated multi-omics approaches, including imaging mass cytometry, single-cell RNA sequencing, Visium and Xenium spatial transcriptomics, and multiplex immunofluorescence. Xenium profiling was performed on FFPE lymph node sections spanning different metastatic stages to resolve high-resolution spatial organisation, neighbourhood complexity, and immune cell co-localisation during sequential lymph node metastasis.

Here, we demonstrate that a bifunctional TGF-βRII/IL-15 protein complex (HCW9218), with TGF-β neutralization and IL-15 immunostimulatory activities, is capable of localizing to the TDLNs and tumors after subcutaneous administration, neutralizes TGF-β, expands progenitor stem-like exhausted T cells (TPEX) in TDLNs, and promotes their infiltration into tumors. Immune-checkpoint-inhibitors (ICIs) significantly enhanced the effects of HCW9218 on TPEX, and synergistically improved HCW9218 anti-tumor efficacy in melanoma and reduced spontaneous lung metastasis in breast cancer models. In a dose-escalating clinical trial in patients with chemo-refractory/relapsed solid tumors, HCW9218 monotherapy was well-tolerated, reduced serum TGF-β levels, promoted CD8+ T cell expansion in peripheral blood and CD8+ T cell infiltration in tumor biopsies. These findings provide a strong basis for using HCW9218 to enhance the efficacy of ICIs against solid tumors in the clinical setting.

Project description:Tumor-draining lymph node (TDLN) invasion by metastatic cells in breast cancer correlates with poor prognosis and is associated with local immunosuppression, which can be partly mediated by regulatory T cells (Tregs). Here, we study Tregs from matched tumor-invaded and non-invaded TDLNs, and breast tumors. We observe that Treg frequencies increase with nodal invasion, that Tregs express higher levels of co-inhibitory/stimulatory receptors than effector cells. Also, while Tregs show conserved suppressive function in TDLN and tumor, conventional T cells (Tconvs) in TDLNs proliferate and produce Th1-inflammatory cytokines, but are dysfunctional in the tumor. We describe a common transcriptomic signature shared by Tregs from tumors and nodes, including CD80, which is significantly associated with poor patient survival. TCR RNA-sequencing analysis indicates trafficking between TDLNs and tumors and ongoing Tconv/Treg conversion. Overall, TDLN Tregs are functional and express a distinct pattern of druggable co-receptors, highlighting their potential as targets for cancer immunotherapy.

Project description:Metastasis to lymph nodes is an early and prognostically important event in the progression of many human cancers, and is associated with expression of vascular endothelial growth factor-D (VEGF-D). Changes to lymph node vasculature occur during metastasis, and may establish a metastatic niche capable of attracting and supporting tumor cells. We used microarrays to characterise the molecular profiles of endothelial cells from lymph nodes draining metastatic (VEGF-D-overexpressing) and non-metastatic tumors, and to identify differentially-expressed genes that might have therapeutic or prognostic potential. Draining lymph nodes of metastatic (VEGF-D-overexpressing) or non-metastatic tumors were pooled from 1-5 mice and enzymatically digested. Lymph nodes draining metastatic tumors were included for the analysis only if macroscopically enlarged, indicating the presence of metastatic cells. After digestion, tumor cells and leukocytes were depleted via immunomagnetic selection, and the resulting lymph node stromal cells were cultured briefly. Podoplanin was then used as a positive immunomagnetic selection marker to enrich for lymphatic and other endothelial cells in the lymph node. RNA was isolated from biological duplicate lymph node endothelial cell (LN EC) preparations and analysed by microarray.

Project description:Tumor-draining lymph nodes (TDLNs) play a crucial role in anti-tumor immunity. However, the heterogeneity of TDLNs significantly impacts their immune function. We employed single-cell RNA sequencing to dissect the immune landscape of TDLNs of colorectal cancer (CRC), revealing that enlarged non-metastatic TDLNs (L-TDLNs) are enriched with CD8+ effector T cells (Teff) exhibiting a distinct metallothionein (MT)-positive signature. These CD8+MTs+ Teff cells, characterized by heightened cytotoxicity and a stress-adapted phenotype, are critical mediators of anti-tumor immunity. Our data indicate that these cells migrate from L-TDLNs to tumor sites, enhancing tumor immunogenicity. Moreover, CRC patients with a high level of CD8+MTs+ Teff in tumor site showed a significant survival advantage, particularly when treated with adjuvant chemotherapy. These findings underscore the importance of L-TDLNs in shaping effective antitumor immune responses in CRC, suggesting that the CD8+MTs+ Teff cell population may serve as a novel prognostic biomarker and therapeutic target. Our study provides a comprehensive framework for understanding the cellular dynamics within TDLNs and their impact on CRC progression and treatment response.

In this study, fresh surgically resected specimens from treatment-naive TNBC patients were collected and CD45+ immune cells were isolated for single-cell RNA sequencing to characterize immune remodeling across matched primary tumors, sentinel tumor-draining lymph nodes (S-TDLNs), and secondary axillary lymph nodes (ALNs). The overall project integrated scRNA-seq with imaging mass cytometry, Visium and Xenium spatial transcriptomics, and multiplex immunofluorescence. This GEO submission contains processed scRNA-seq expression matrices and associated metadata only. Raw sequencing data have been deposited previously in in the Genome Sequence Archive (GSA) for human data, and the GSA ID is HRA010347.
