Single-cell transcriptomic analysis of Ikaros and/or Helios deficient HSPCs

ABSTRACT: Here we addressed the single cell transcriptome of hematopoietic stem and progenitor cells (HSPCs/LSK) purified from WT (Ikzf1+/+ Ikzf2+/+ Rosa26CreERT2/+), Helios KO (Ikzf2F/F Rosa26 CreERT2/+), Ikaros KO (Ikzf1F/F Rosa26 CreERT2/+) and DKO (Ikzf1F/F Ikzf2F/F Rosa26CreERT2/+) mice, 5 days after the last tamoxifen injection used to induce the deletion of Ikaros and/or Helios. We used single cell RNA sequencing (scRNA-seq) to determine the function of Ikaros and Helios in modulating the HSPCs lineage commitment and to study how their loss affect the composition of the HSPC compartment in terms of cell subtypes.
