Single-cell RNA-seq of immune and stromal cells from murine muscle injury and foreign body response to synthetic biomaterial implants

ABSTRACT: The main objective of this study is to characterize the immune and stromal cell response to tissue injury and the foreign body response (synthetic polycaprolactone PCL implant). We enrich CD45+ immune cells from injured murine muscles with fibrotic PCL implants (6 week time point) and perform transcriptomic profile using single-cell RNA-sequencing.

We isolate key stromal cell populations (Fibroblasts and Endothelial Cells) from fibrotic PCL implants of wildtype (WT) and GD-deficient (GDKO) mice using FACS and compare their transcriptomic profile using bulk RNA-sequencing.

Murine gut microbiota dysbiosis via enteric infection modulates the foreign body response to a distal biomaterial implant

Project description:The gut microbiota influences systemic immunity and the function of distal tissues, including the brain, liver, skin, lung, and muscle. However, the role of the gut microbiota in the foreign body response (FBR) and fibrosis is largely unexplored. To investigate this connection, we perturbed the homeostasis of the murine gut microbiota via infection with the pathogenic bacterial species enterotoxigenic Bacteroides fragilis (ETBF) and implanted particulate material (mean particle size <600 μm) of the synthetic polymer polycaprolactone (PCL) into a distal muscle injury. ETBF infection in mice led to increased neutrophil and γδ T cell infiltration into the PCL implant site. ETBF infection alone promoted systemic inflammation, increased levels of neutrophils in lymphoid tissues, and altered skeletal muscle gene expression. At the PCL implant site, we found significant changes in the transcriptome of sorted stromal cells between infected and control mice, including differences related to ECM components such as proteoglycans and glycosaminoglycans. However, we did not observe ETBF-induced differences in fibrosis levels. These results demonstrate the ability of the gut microbiota to mediate long-distance effects such as immune and stromal responses to a distal biomaterial implant.

Single-cell Analysis of Surface Topography-induced Foreign Body Response to Silicone Implants

Project description:Silicone-based medical devices are widely used in chronic implants and are generally perceived to be safe. However, immune-related complications including malignancies have recently been linked to textured breast implants. Here, we examine the influence of clinically approved breast implants surface features on host immune responses. Prosthetics with surface roughness of 0, 4, and 90 (Ra) were implanted in mammary fat pads of mice for 2 weeks and cells adjacent to the resulting tissue capsules were evaluated for foreign body immune responses using single-cell RNA-seq. Our findings identify a unique and finely tuned surface topography that is capable of modulating implant immunity to suppress foreign body response.

Engineered biomaterial immunological niches as a surrogate for diseased tissue in chronic progressive EAE

Project description:We use a subcutaneous biomaterial implant as a surrogate for sampling immune cells phenotypes in murine chronic EAE. These implants are microporous polycaprolactone scaffolds that form immunological niches that get vascularized and capture both innate and adaptive immune cells. These niches can be minimally invasively biopsied, allowing for multiple samples to be collected from the same mouse at various timepoints in disease: presymptomatic (Day 7) and early state disease (Day 9). This study investigates immune dynamics over time in subclinical and early stage chronic progressive EAE at a distal biomaterial-based immunological niche.

Osteopontin Reduces Foreign Body Response in Silicone-Based Implants in Humans and Mice

Project description:Subcutaneous implants often elicit an inflammatory foreign body response (FBR) in the shape of a capsule around the implanted material, and it is not yet clear why certain materials such as dermal matrix (ADM), clinically used in breast reconstruction, are more efficient in attenuating this chronic reaction. Here, we investigate the molecular response of human and mouse tissues implanted with silicone treated with ADM by analysing single-cell RNA and proteomics. In patient-derived implanted tissues exposed to ADM, we identify high levels of Osteopontin-1 (SPP1) together with attenuated FBR, in contrast to higher inflammatory response in implanted tissues not exposed to ADM. In a Spp1 knockout mouse model of FBR, we demonstrate that a reduction in SPP1 expression prevents the beneficial effects of ADM in the capsule formation, revealing that the protein is necessary to decrease fibrotic response. In wildtype mice, we demonstrate that a sustained release from a hydrogel of recombinant Spp1 is sufficient to reduce fibrotic encapsulation even in the absence of ADM, suggesting a therapeutic approach for reducing FBR around implants.

Degradations of different processed Mg alloy implants lead to distinct foreign body reactions (FBRs)

Project description:Mg alloy is one of the lightest metals on earth and has the most similar mechanical properties to human bones. However, the active chemical properties and fast in vivo degrade speed significantly hinder its further employment as human implant. In this study, three different Mg alloys are employed to investigate the in vitro corrosion behavior including weight loss, pH value evolution and surface hardness and in vivo degradable speed and tissue compatibility. All three Mg alloys are well tolerated when implanted in SD rats, with minimal influence on hepato-and renal functions and vital organs. Compared to cold extruded AZ31 (CE AZ31) and pure Mg (PM) specimens, fully annealed AZ31 (FA AZ31) presents much stable surface asperity. Proteomics analysis of tissues near implant site showed that FA AZ31 activates few inflammation and immune associated signaling pathways; while the CE AZ31 and pure Mg led more significant inflammatory responses as confirmed by the cytokine array that pure Mg stimulate higher IL-1 production than FA AZ31. Further, FA AZ31 can activate pathways of cell organization and development that may improve the recovery of the injured tissues neighboring the implant sites. Besides, all three Mg alloys cannot inhibit a methicillin-resistant S. aureus growth if the implants are contaminated with this bacterium. FA AZ31 has a higher ratio of first-order pyramidal slips system (10-11) {10-1-2} than pure Mg and cold extruded AZ31 analysed by EBSD, suggesting the process of dynamic recovery may plays an important role in the improvement of mechanical properties, especially bio-properties including corrosion resistance and biocompatibility. In conclusion, FA AZ31 has better biocompatibilities and corrosion resistance, makes it a promising candidate of metal-based degradable implant and warrant further investigation.

Metastatic conditioning of myeloid cells at a subcutaneous synthetic niche reflects disease progression and predicts therapeutic outcomes

Project description:Microporous polymer (PCL) implants were implanted into BALB/c mice. After 2 weeks mice were inoculated with 4T1 tumor cells. Then after 7, 14, and 21 days the implant and tissue infiltrate which develops into a synthetic niche was biopsied, total RNA was isolated, reverse transcribed into cDNA and analyzed using a high throughput RT-qPCR platform (Applied Biosystems™ TaqMan™ OpenArray™ Mouse Inflammation Panel). The goal of the studies was to identify genes that progressively change at a synthetic site during disease progression.
