Human ZBP1 Is a Potent Inducer of RIPK1-Dependent Cell Death Through Mechanisms Divergent from Mouse ZBP1 [mouse]


ABSTRACT: Z-DNA-binding protein 1 (ZBP1) is a key mediator of innate immunity, sensing Z-form nucleic acids (Z-NAs) via two Zα domains and initiating cell death and inflammation through three RHIM domains. While the regulatory mechanisms of mouse ZBP1 (mZBP1) have been well studied, the pathways governing human ZBP1 (hZBP1) remain poorly understood. In this study, we show that unlike mZBP1, overexpression of hZBP1 induces spontaneous cell death in human cell lines. This response involves activation of both FADD–caspase-8-mediated apoptosis and RIPK3–MLKL-dependent necroptosis. Notably, in contrast to mZBP1, which signals primarily through RIPK3, hZBP1-induced cell death depends on the scaffold function of RIPK1 but not its kinase activity. Importantly, this hZBP1-RIPK1 engagement occurs independently of RIPK3. Using chimeric ZBP1 constructs, we identified the C-terminal region containing RHIM2 and RHIM3 as the key determinant that drives spontaneous cell death upon hZBP1 overexpression. We also confirmed that endogenous hZBP1 promotes RIPK1-dependent cell death under pathological conditions, including ADAR1 deficiency and treatment with specific drugs. Together, our findings reveal an intrinsic mechanistic divergence between human and murine ZBP1 signaling and highlight the limitations of translating preclinical findings in animal models to human therapeutic strategies targeting this pathway.
