SPP1+ Macrophages Contribute to Hair Growth in Contact Hypersensitivity [scRNA-Seq]

ABSTRACT: Skin inflammation involves complex interactions between immune cells. To address this gap, we applied single-cell RNA sequencing to dissect cellular states and regulatory programs in DPCP-induced dermatitis. This approach enables high-resolution mapping of immune crosstalk and identification of novel macrophage subsets. Our study establishes a framework for understanding how immune and metabolic signals orchestrate skin inflammation and regeneration.

Project description:Using Visium HD spatial transcriptomics and immunostaining, we mapped skin compartment dynamics during DPCP-induced allergic contact dermatitis. Dermal white adipose tissue transitioned from metabolic homeostasis to immune activation and inflammatory remodeling, accompanied by sustained lipolysis. Concurrently, CD45⁺ immune cells accumulated in the lower dermis and dWAT. These findings identify dWAT as an immune-metabolic hub orchestrating inflammatory progression and highlight spatial transcriptomics as a powerful tool for dissecting skin niche regulation.

Unique microRNAs appear at different times during the course of a delayed-type hypersensitivity reaction in human skin

Project description:Diphencyprone (DPCP) is a hapten that induces delayed-type hypersensitivity (DTH) reactions. MicroRNAs (miRNAs) are short non-coding RNAs that negatively regulate gene expression, and have been implicated in various inflammatory skin diseases, but their role in DTH reactions is not well understood. We generated global miRNA expression profiles (using next-generation sequencing) of DPCP reactions in skin of 7 healthy volunteers at 3, 14, and 120 days after challenge. Compared to placebo-treated sites, DPCP-challenged skin at 3 days (peak inflammation) had 127 miRNAs significantly deregulated. At 14 days (during resolution of inflammation), 43 miRNAs were deregulated and, at 120 days (when inflammation had completely resolved), 6 miRNAs were upregulated. While some miRNAs have been observed in psoriasis or atopic dermatitis, most of the deregulated miRNAs have not yet been studied in the context of skin biology or immunology. Across the three time points studied, many but not all miRNAs were uniquely expressed. As various miRNAs may influence T cell activation, this may indicate that the miRNAs exclusively expressed at different time points function to promote or resolve skin inflammation, and therefore may inform on the paradoxical ability of DPCP to treat both autoimmune conditions (alopecia areata) and conditions of ineffective immunity (melanoma).

Transcription profiling of mouse Cannabinoid receptor double knockouts (Cnr1 -/- /Cnr2 -/-) in CHS model to investigate cutaneous contact hypersensitivity

Project description:We evaluated cutaneous contact hypersensitivity (CHS) in Cnr1-/-/Cnr2-/- animals using the obligate contact allergen 2,4-dinitrofluorobenzene (DNFB), which generates a specific cutaneous T-cell mediated allergic response upon repeated allergen contact. Allergic contact dermatitis affects about 5% of men and 11% of women in industrialized countries and is one of the leading causes for occupational diseases. In an animal model for cutaneous contact hypersensitivity we show that mice lacking both known cannabinoid receptors display exacerbated allergic inflammation. In contrast, fatty acid amide hydrolase deficient mice, which have increased levels of the endocannabinoid anandamide, displayed reduced allergic responses in the skin. Cannabinoid receptor antagonists exacerbated whereas receptor agonists attenuated allergic inflammation. These results demonstrate a protective role of the endocannabinoid system in contact allergy in the skin, and suggest a novel target for therapeutic intervention. Experiment Overall Design: Three wildtype mice (Wt) and three Cnr1-/-/Cnr2-/- (Ko) mice were used. Contact hypersensitivity was determined always at the right ears, which therefore were treated with DNFB (Tr). Left ears of mice were kept untreated and served as control ears (C). A total of 12 hybridizations were performed (2 strains x 2 treatments X 3 biological replicates) in this experiment.
