Z-RNA drives Tau-mediated neurodegeneration of Alzheimer’s disease [RNA-Seq]

ABSTRACT: Tau aggregates lead to progressive neurodegeneration in Alzheimer’s disease (AD) (ref). Neuron death is one of the hallmarks of neurodegeneration (ref). However, the pathological influence of neuronal death is undetermined, and the connection between Tau aggregates and neuronal death remains elusive. Here we demonstrated the essential role of neuron death in Tau-related neurodegeneration. Tau-neurons died in necroptosis, dependent on ZBP1 sensitized by Z-RNAs (an unusual left-handed conformation). Those endogenous Z-RNAs were transcripts of reactivated transposable elements (TEs) originally silenced in heterochromatin.

Spliceosome inhibition induces Z-RNA and ZBP1-driven Cell Death in Small Cell Lung Cancer

Project description:Spliceosome inhibitors emerged as promising anticancer agents. Recent studies have demonstrated that spliceosome targeted-therapies (STTs) trigger antitumor immune responses by inducing the accumulation of right-handed double-stranded (ds)RNA (A-RNA), resulting in activation of RIG-I-like receptors and Type I interferon-driven antiviral responses. Here, we show that pharmacological spliceosome inhibition or genetic neutralization of SF3B1 activity induces the accumulation of endogenous left-handed dsRNAs (Z-RNAs) derived from intron-retained RNAs. These Z-RNAs activate the Z-form nucleic acid-sensor ZBP1, which triggers cell death in mouse embryonic fibroblasts (MEFs) and small cell lung cancer (SCLC) cells. Spliceosome inhibition induced potent ZBP1-dependent cell death in cancer-associated fibroblasts (CAFs), which was essential for enhancing immunotherapy response in mouse models of SCLC. Collectively, these results demonstrate that spliceosome inhibitors or STTs, can be used to generate Z-RNA and trigger on-demand ZBP1-dependent cell death in cells of the tumor microenvironment (TME) as a therapeutic strategy to enhance immunotherapy responses in resistant-cancers.

Casein kinase 1δ induces the formation of GVB-positive neurons that are resilient to tau-induced impairment of protein synthesis

Project description:The progressive intraneuronal accumulation of protein aggregates is central in the pathogenesis of several neurodegenerative diseases. Therefore, molecular pathways that mediate neuronal resilience to tau pathology present interesting targets for therapeutic intervention. To this end we investigated the mechanism and functional implications of the formation of granulovacuolar degeneration bodies (GVBs), neuron-specific lysosomal structures that form in response to pathological tau assemblies. In the present work, we show that intraneuronal tau pathology progressively induces the formation of GVB+ neurons. We demonstrate that CK1δ activity is a rate limiting step in the formation of GVB+ neurons that requires the autophagic machinery. The GVB neuronal state confers protection against tau pathology: GVB+ neurons have structural adaptations in lysosomal and ribosomal content to become resilient to tau-induced protein synthesis impairment. Moreover, GVB+ neurons are functionally resilient to tau-induced neurodegeneration and impairment of LTP-regulated protein synthesis. Altogether our results have identified CK1δ as a key regulator of a protective neuron-specific proteostatic response to tau pathology.

Neuronal identity defines a-synuclein and tau toxicity

Project description:Pathogenic a-synuclein and tau are critical drivers of neurodegeneration and their mutations cause neuronal loss in patients. Whether the underlying preferential neuronal vulnerability is a cell-type intrinsic property or a consequence of increased expression levels remains elusive. Here, we explore cell-type specific a-synuclein and tau expression in human brain datasets and use deep phenotyping as well as brain- wide single-cell RNA sequencing of >200 live neuron types in fruit flies to ask which cellular environments react most to a-synuclein or tau toxicity. We detect phenotypic and transcriptomic evidence of differential neuronal vulnerability independent of a-synuclein or tau expression levels. Comparing vulnerable with resilient neurons in Drosophila enabled us to predict numerous human neuron subtypes with increased intrinsic susceptibility to pathogenic a-synuclein or tau. By uncovering synapse and Ca2+ homeostasis related genes as tau toxicity modifiers our work paves the way to leverage neuronal identity to uncover modifiers of neurodegeneration-associated toxic proteins.

KCTD20 suppression mitigates excitotoxicity in tauopathy patient organoids

Project description:Excitotoxicity is a major pathologic mechanism in patients with tauopathy and other neurodegenerative diseases. However, the key neurotoxic drivers and most effective strategies for mitigating these degenerative processes are unclear. Here, we show that glutamate treatment of induced pluripotent stem cell (iPSC)-derived cerebral organoids induces tau oligomerization and neurodegeneration, and that these phenotypes are enhanced in organoids derived from tauopathy patients. Using a genome-wide CRISPR interference (CRISPRi) screen, we find that suppression of KCTD20 potently ameliorates tau pathology and neurodegeneration in glutamate-treated organoids and mice, as well as in transgenic mice overexpressing mutant human tau. KCTD20 suppression reduces oligomeric tau and improves neuron survival by activating lysosomal exocytosis, which clears pathological tau. Our results show that glutamate signaling can induce neuronal tau pathology and identify KCTD20 suppression and lysosomal exocytosis as effective strategies for clearing neurotoxic tau species.
