Exploring HDAC8 related transcriptomic change in DNA damage response in AML

ABSTRACT: To explore the underlying changes during the DNA damage in HDAC8-ovexpressing AML, we performed RNA-seq for MV4-11 cells overexpressing HDAC8 (MV4-11-HDAC8) and empty vector controls (MV4-11-EV) at 12h after irradiation (IR) or no IR (NIR).

Project description:Analysis of the RNA-seq data performed in IR vs NIR hematopoietic stem cells show the loss of the TNF_via_NFKB signature. We showed that the loss of this signature could be associated with hematopoietic stem cell gene deregulation. To validate this association, we tested if TNFa treatment before irradiation was able to prevent IR-effect on HSC gene deregulation. For this purpose, we treated mice with TNFa 1h before irradiation (IR_TNF) and performed RNA-seq experiments on hematopoietic stem cells 1 month after irradiation and compared them to hematopoietic stem cells sorted from non irradiated mice (NIR) and from non-treated irradiated mice (IR).

Infrared light enhances deep-sea yeast growth through temperature insensitive wavelength-dependent mitochondrial metabolism

Project description:Deep-sea hydrothermal vents emit intense infrared (IR) radiation that profoundly shapes microbial ecology and promotes bacterial growth. To investigate the mechanisms underlying IR responsiveness, we isolated Rhodotorula mucilaginosa strain F-J1, a deep-sea yeast. We found that both elevated temperature and near-infrared (NIR, 810–1050 nm) irradiation stimulated F-J1 growth. Whereas dark-grown cultures were highly temperature-sensitive, NIR irradiation—particularly at 880 nm—not only enhanced growth but also substantially lowered the Arrhenius activation energy, indicating a wavelength-dependent, temperature-insensitive stimulation mechanism. At the atomic level, substituting hydrogen with deuterium (via heavy water) suppressed F-J1 growth. This inhibition was partially alleviated by wavelength-dependent NIR irradiation, implicating water’s overtone vibrations. However, deuterium completely abolished the stimulatory effect of 880 nm NIR, revealing a distinct, wavelength-specific mechanism. Inhibition of mitochondrial ribosome function and oxidative phosphorylation impaired NIR-stimulated growth, underscoring the critical role of mitochondrial metabolism. Electron microscopy further showed NIR-induced mitochondrial morphological changes consistent with heightened metabolic activity. Proteomic analysis identified upregulation of mitochondrial ribosomal protein S18, suggesting enhanced mitochondrial protein biosynthesis under optimal NIR conditions. Together, these findings uncover a mitochondria-centered, non-thermal IR-sensing mechanism that parallels known photobiomodulation processes in mammalian cells. This work provides new insights into the ecological role of IR radiation in deep-sea hydrothermal ecosystems and highlights its potential biomedical and biotechnological applications.

Effect of mutation at the K202 site of the HDAC8 on 3D genomic structure in HeLa cells

Project description:HDAC8, a member of class I HDACs, plays a pivotal role in cell cycle regulation by deacetylating the cohesin subunit SMC3. While cyclins and CDKs are well-established cell cycle regulators, our knowledge of other regulators remains limited. Here we reveal the acetylation of K202 in HDAC8 as a key cell cycle regulator responsive to stress. K202 acetylation in HDAC8, primarily catalyzed by Tip60, restricts HDAC8 activity, leading to increased SMC3 acetylation and cell cycle arrest. Furthermore, cells expressing the mutant form of HDAC8 mimicking K202 acetylation display significant alterations in gene expression, potentially linked to changes in 3D genome structure, including enhanced chromatid loop interactions. K202 acetylation impairs cell cycle progression by disrupting the expression of cell cycle-related genes and sister chromatid cohesion, resulting in G2/M phase arrest. These findings indicate the reversible acetylation of HDAC8 as a cell cycle regulator, expanding our understanding of stress-responsive cell cycle dynamics.
