The difference of gene expression and splicing between the WT and scl mutants.

ABSTRACT: Transcriptional and post-transcriptional regulation are crucial for plant development and stress responses. We here report the SR gene as a key regulator of gene expression and alternative splicing in rice (Oryza sativa). Comparative RNA-seq analysis between wild-type and scl mutant plants revealed significant differential expression of genes involved in growth, development pathways. Notably, the sr mutation resulted in widespread alterations in alternative splicing patterns, particularly affecting genes involved in signal transduction and transcriptional regulation. Although the sr mutation did not affect the binding of general transcription factors to promoter regions, it specifically impaired the coordination between transcription and splicing machinery for a subset of genes with critical developmental functions. Taken together, our data provide a molecular framework for understanding how the scl gene integrates transcriptional and splicing regulation in rice, offering new insights into the mechanisms governing plant development and potential targets for crop improvement.
