Project description:Background:HuR/ELAV1, a ubiquitous RNA-binding protein, belongs to the RNA-binding protein family and is crucial for stabilizing and regulating the translation of various mRNA targets, influencing gene expression. Elevated HuR levels are associated with multiple disorders, including cancer and neurodegenerative diseases. Despite the identification of small molecule inhibitors targeting HuR, their detailed characterization remains limited. Recently, Eltrombopag, an FDA-approved drug for immune thrombocytopenic purpura and chemotherapy-induced thrombocytopenia, emerged as a potential HuR inhibitor. However, the specific molecular pathways influenced by both HuR and Eltrombopag are not fully understood. Results:Our study demonstrates that Eltrombopag operates via HuR inhibition, affecting gene expression regulation at the posttranscriptional level. We show that both HuR knockout and Eltrombopag treatment modulate iron metabolism by decreasing ferritin heavy chain (FTH1) and light chain (FTL) synthesis while increasing the expression of Iron-regulatory protein 2 (IRP2), a key regulator of ferritin translation. Additionally, HuR inhibition reduces the levels of Glycoprotein Hormones, Alpha Polypeptide (CGA), a marker associated with hormone-induced tumors, suggesting a potential use of Eltrombopag in treatment of cancers overexpressing CGA. We observed that the main of control is manifested at the level of translation inhibition, with proteosome-mediated regulation also playing an important role.Conclusions:These findings uncover novel posttranscriptional mechanisms governed by HuR and its inhibitor, elucidating pathways relevant to HuR-mediated regulation and molecular therapies aimed at targeting this protein. Project description:Mutations in isocitrate dehydrogenase 1 (IDH1-R132H) co-occur with TP53 and ATRX loss in adult low-grade glioma. Using a genetically engineered mouse glioma model (Sleeping Beauty transposon system; NRAS(G12V), shRNA-mediated Trp53 and Atrx knockdown, with or without mutant IDH1-R132H) and derived neurosphere (NS) cultures, we asked whether IDH1-R132H alters nascent transcription of DNA damage response (DDR) genes. Nascent RNA was metabolically labeled with 5-bromouridine (Bru) and Bru-labeled transcripts were immunopurified and sequenced (Bru-seq) to measure genome-wide nascent transcription rates at baseline (untreated), independent of steady-state mRNA abundance. Bru-seq identified increased nascent transcription of DDR genes, including Atm and Rad50, in IDH1-R132H-mutant (mIDH1) NS relative to wild-type IDH1 (wt-IDH1) NS, consistent with the epigenetic up-regulation of ATM signaling and enhanced DDR reported for this model.

The aim of the project was to characterize the role of HSPA2, a member of the HSPA chaperone family (HSP70), in human epidermal keratinocytes. The inactivation of the HSPA2 gene in the HaCaT line of spontaneously immortalized epidermal keratinocytes was performed by CRISPR/Cas9 gene editing system. Next, the effect of modifications on the transcriptomic profile of cells growing in 2D monolayer culture was investigated. Project description:Animal models provide opportunity to study neurobiological aspects of human alcoholism. Changes in gene expression have been implicated in mediating brain function, including reward system and addiction. The current study aimed to identify novel genes that may underlie ethanol preference. Microarray analysis comparing gene expression in nucleus accumbens (NAc), hippocampus (HP) and prefrontal medial cortex (mPFC) was performed in two rat strains selected for extreme levels of ethanol preference - Warsaw High Preferring (WHP) and Warsaw Low Preferring (WLP). The identified candidate genes may underlie differential ethanol preference in rat model of alcoholism. The cells were grown at air-liquid interface culture on extracellular matrix to achieve maximal level of HaCaT differentiation in RHE system. Project description:Advanced Maternal Age (AMA) is a risk factor for neurological and neuropsychiatric disorders in offspring. We developed a mouse model to investigate whether pregnancy at advanced age may provoke behavioral and brain gene expression changes in offspring. Mice conceived by 15- to 18-month-old (15-18M) or 3M control females were delivered by cesarean section and were all fostered after birth by 3M dams. Genome-wide mRNA expression was analyzed using microarrays in the hippocampus of 4M male offspring. In conclusion, pregnancy at advanced age yields offspring with altered patterns of gene expression in the hippocampus, typical of human neuropsychiatric and neurodegenerative disorders. The effects were not reversed by the postnatal maternal care provided by young foster mothers. This suggests that prenatal conditions associated with AMA may negatively affect fetal brain development and hence postnatal behaviors.
