Migrating adipose stromal cells fuel regenerative adipogenesis in injured muscle

ABSTRACT: Muscle repair following injury is a complex process. We previously demonstrated the key role played by Adipose stromal cells (ASCs) from subcutaneous adipose tissue in this process. Using single-cell RNA-seq approach we were able to trace which ASC sub-populations contribute to this phenomenon. In addition following these cells in the muscle, we propose that their transient differentiation into adipocytes is beneficial to muscle repair.

Small RNA-seq of small extracellular vesicles derived from adipose-derived stem cells of young and old donors at different in vitro expansion stages

Project description:Adipose-derived stem cells (ASCs) and their small extracellular vesicles (sEVs) hold significant potential for regenerative medicine due to their capabilities for tissue repair. However, the effects of ASC heterogeneity on sEV characteristics and microRNA (miRNA) profiles remain unclear. We investigated ASCs heterogeneity and its changes with aging and cell expansion using bulk RNA-seq from young and old donors across various passages. Non-negative matrix factorization (NMF) was used to identify distinct ASC clusters. Small RNA sequencing was performed on sEVs collected from ASC culture supernatants to analyze miRNA profiles. NMF analysis revealed ASC heterogeneity, identifying clusters characterized by ISLR and genes associated with collagen and smooth muscle cells. The miRNA profiles of sEVs matched these ASC clusters, showing variations based on donor age and cell expansion. Functional enrichment analysis suggested that these miRNAs may have distinct functional roles. Our findings show that ASC heterogeneity and sEV miRNA content are significantly influenced by donor age and cell expansion. These variations underscore the necessity of selecting specific ASC-derived sEVs.

Project description:Muscle Stem Cells or satellite cells (SCs) are required for muscle regeneration. In resting muscles, SCs are kept in quiescence. After injury, SCs undergo rapid activation, proliferation and differentiation to repair damaged muscles. The transcriptome alteration during SC activation is well characterized. While transcriptome is not exactly represent proteome because of post-transcriptional regulations such as miRNA induced gene silencing. However, little is known about SC proteome. We obtained in vivo activated SCs (ASCs) from 3 days post injured muscles for high resolution mass spectrometry Bruker timsTOF Pro. Compared with QSC proteome,, we identified the pathways that are differentially expressed between them.

Project description:There is a large body of evidence supporting the beneficial role of adipose-derived stem cells (ASCs) in tissue repair and regeneration. These effects appear to be mainly mediated by paracrine signaling pathways and enhanced during hypoxia. Mass spectrometry (MS) represents a valuable tool for proteomic profiling of cultured ASCs, which may help revealing the identity of the factors secreted by the cells under different conditions. However, serum starvation essentially required to obtain samples compatible with secretome analysis by MS can have significant influence on ASCs. Here, we present a novel and optimized culturing approach based on the use of a clinically relevant serum-free formulation, which was used to assess the effect of hypoxia on the ASC proteomic profile.
